very easy and informative video, you have explained it very well, thank you ma'am, but no explanation about the addition of ladder and positive control and negative control, plz tell about these too ?

A DNA ladder is just a standard set of DNA fragments that are various, but known, lengths (bought from a biological supply company). They will be pulled through the gel, with the smallest pulled through faster than the larger fragments. When the gel is disconnected and stained, the ladder can be compared to the unknown fragments in other wells/columns, so their lengths can be estimated. (The ladder is used to make a standard curve with semi-log paper (distance vs. length of fragment), and the unknown fragments can be estimated by using the curve made with the semi-log paper). I don’t think we actually used a ladder or controls for this lab, just the three strains of (fake) virus DNA (it's a 9th grade, intro to biotechnology lab).
