But according to this method you cannot go to 0 after 2,1,0 as it's a match (G and G) so you have to go diagonally, which is 2,1,0,-1. Note: here you can't go according to the highest value if the nucleotide sequence matches. If it doesn't match then you can go for the highest value.

Because this is the 1st rule. Before filling up a matrix you have to fill the first row and column by adding the given score of a gap penalty (which is -2 here) with 0. And the first one will always have to be 0.

hi I may be doing it wrong but could you please confirm for me. for the third example, the highest available value in the matrix filling is 4 and it is not from the very bottom right. do I still start from the very bottom right then? or from the highest value? if I start from the highest value available my answer is seq 1: C G CTCGCT seq 2: C(A)CTCG - As seen here there is a mismatch, where the brackets represents it and the sequence is cut off by one nucleotide at the back if I start from the very bottom right regardlessly, seq 1: C G CTCGCT seq 2: C(A)CTC_CT , once against bracket is mismatch present Which would be the right method please? also can I confirm my answers for the rest of the try it questions too please: Seq1 : AATCG Seq 2: AA_CG Seq 1: CCGTCG Seq 2: CCG_CG Thank you so much.

the cost for changing sequence a to b is 2 so there must be two changes but in 10:54 there is only one change so doesn't it become as a naïve solution?
