Hi Jesse! I was wondering if you could tell me why they seem to buddy paracetamol up with a lot of other pain relieving drugs such as, Ibuprofen, Codeine, Diazepam or Tylenol.. this seems like pain-relief overkill

Yeah absolutely! So pain relief drugs can often be more effective when used in conjunction with one another, and it gets pretty complicated! Tylenol is the American version of Paracetamol (Panadol), and when used in conjunction with ibuprofen, they work together to COX-BLOX pain by getting rid of the prostaglandins that cause pain and fever. Opiate drugs like morphine and codeine work directly on the nervous system to reduce pain signals all together. And Diazepam is a relaxant that is usually prescribed to calm you down and make you sleepy...maybe to sleep through the pain. I will have to make a video that discusses how various drugs interact together when they are used at the same time. Great Question Chalie!

@@TheTravellingScientist Hey there, great video. I have CRPS and I've been treated by some of the best specialists around here. I'm like 10% of the gen pop that can't turn codeine into morphine which happens in the liver I think. The diazepam (valium) is the absolute best at releasing a spasm. They work better than any other muscle relaxer but only at small dosages. Certain Antidepressants help the analgesic work better, so most people with any chronic pain illness usually adds this to their regiment. Your videos are very informative so thank you.

It is possible that paracetamol could have some impact on the neurotransmitters in your brain like noradrenaline, which could alter your memory slightly. This is a rare but annoying side effect. I hope it doesn't affect you too severely

That is a great question! So paracetamol was first synthesized over 140 years ago, so it was poorly understood, but it was recognized as an effective pain reliever. 140 years later and not much had changed, haha

@@ivan0kurnia Well decades ago when the drug was first produced, you didn't need volunteers. They would test drugs like these on prison inmates to work out any side effects. The prisoners had a choice, but they would get paid (minimally) to take novel drugs for testing unknown drugs like Paracetamol

Does paracetamol help mental pain in any way? Like if you felt anxious would taking it make you any less anxious?

Great question! Paracetamol should have little effect on mental pain, it has a purely physical response. However, a reduction in pain would probably improve your mental state, thereby making you feel slightly more happy and less anxious. So indirectly I guess so. I wouldn't prescribe paracetamol for anxiety, but theoretically it could help in this manner. Thanks for getting in touch legend!

ive been suffering for 2 days from a Razor blade stuck in my throat feeling, 8/10 PAIN-SCALE, its been super painful to swallow any food and drink i cant even sleep its soo painful! thats exactly what it feels like a Razor blade stuck in their! OUCH! i gave up, i took 2x 500mg paracitamol about 2 hours ago the pain is still there but heavily reduced i feel loads better than i did 3/10 painscale :O GREAT this is way more comfortable to cope with.

Excellent question @insyko the bear there are many different types of prostaglandins, and they have different effects. In general, they cause pain and inflammation, but some are more pain oriented, while others are more likely to promote swelling, redness, itchiness, etc. Paracetamol seems to have a stronger effect on the pain promoting prostaglandins...gee, try saying that out loud :D Thanks so much for watching and commenting legend!

The paracetamol SHOULD treat your headache! I am looking at making a video about more natural pain treatments, but in the meantime I can tell you that valerian is proven to help people sleep better, but not necessarily to reduce pain from headaches. Willow bark is what aspirin is made out of, and aspirin is great for treating headaches, but willow bark is not as effective, probably more of a placebo effect. Quick question, am training in the pre-hospital environment and a paramedic mentioned that paracetamol and morphine make a good combination is there a reason for this is it to do with the cox 1,2, and 3 I know paracetamol is thought to work on the cox 3.

Great question Gareth, I would have to assume that they make a good combination because they both reduce pain via different pathways. Morphine acts on opioid receptors to reduce pain signal transmission to the spinal cord, while paracetamol acts on Cox to reduce prostaglandin synthesis and inflammation overall. Together, they inhibit almost all pain in the body. Hope that makes sense, thanks for watching!

The drug is not addictive, but if you are suffering from chronic pain, then the pain relief offered by paracetamol can be addictive. It is important to find proper pain solutions that simply covering up pain with a drug like paracetamol, ibuprofen or aspirin! Great question Soha!

Hey @Shubham Agye great question. Paracetamol doesn't really reduce swelling. Ibuprofen would be a better temporary treatment in this case. Can paracetamol reduce covid immune response? I took one after my shot to reduce side effects of the vaccine but now I'm worried if paracetamol will reduce my covid immune response. Is my body reacting the same with or without paracetamol?

Great question! I didn't want to make the video too complicated, but there are 3 different COX enzymes, COX-1, 2 & 3. COX-1 is always produced by your cells, while COX-2 is only produced during periods of stress (injury or disease). COX-3 is a variant of COX-1, almost identical, but it is found mainly in the brain. It is thought that because paracetamol is able to cross the blood brain barrier, it could be acting on COX-3 in the brain to reduce the pain of a headache or similar...but this is still yet to be proven

So, if fever is a natural response of our body to fight against any allergy or problem doesn't taking paracetamol puts us in danger for the long run? Like sure, it will take down my fever but it isn't gonna cure the root problem that caused the fever in the first place and now with paracetamol it also hindered my ability to treat that root cause as well.

Hey legend, did you watch the video? Paracetamol blocks arachadonic acid, thereby preventing the production of prostaglandins which would normally cause pain (headache) and fever (high temperature). "If my temp isn't high then it means that my body isn't fighting infection" is incorrect, that would suggest that this simple tablet is shutting down your immune system...which it isn't. Paracetamol helps us to deal with pain and fever, whether it is caused by an infection or an injury. Either way, our body still works to heal the infection or injury, but if we don't feel okay, we can take a paracetamol to help us through the pain. Like Asprin and Ibuprofen, Paracetamol is able to cross the placenta. However it has not been shown to cause any adverse effects in pregnant women when the correct dosage is used. So yes, your wife should be fine to use paracetamol if she is in pain :)

Co-Codmal is a combination of codeine and paracetamol and is a more effective pain relief drug than paracetamol or aspirin. If you are taking this drug WITH Iburpofen and are still in pain, something is wrong. Chronic pain is a very common medical issue. I would recommend discussing the pain with your doctor and finding an appropriate treatment, perhaps you need some sort of massage, physiotherapy, a chiropractor or even some sort of surgery. Unfortunately there is no way to measure concentrations of anything cheaply, and plant sources can very greatly dependent on the growth situation. The best thing you can do is to find a reference concentration and then roughly estimate using that reference knowledge. Does that make sense? Sorry I can't help more

Bir 3 paracetamol at one time is okay, and would offer effective pain relief. Taking 3 paracetamol regularly is not healthy, it can be taxing on your liver, and she should see a doctor to find the cause of pain rather than keep taking the drug!

Yes, most of it just came to my head when I was learning about it for my job. Then I just filled in the blanks. I find it's easy to remember things when they rhyme and are funny.

So after you take paracetamol, it is quickly absorbed through your stomach wall into your blood stream, where it spreads out and binds to red blood cells. This carries the drug throughout the body where it interacts with cells, inhibiting COX to prevent prostaglandin synthesis and reduce pain levels. Paracetamol is eventually metabolised as it passes through the liver, then it is excreted by our body. Does that answer your question?

That is what I referred to as the "COX-Block", and unfortunately the answer to your question is not certain. I would assume that paracetamol binds to the COX enzyme, inhibiting it's active site and preventing it from interacting with arachidonic acid to produce painful prostaglandins, but that is not certainly how it works...that is just a guess, nobody knows for certain how this drug works, that is a key point of this video. Crazy huh!?

Could this be the answer to why it doesn't work for pain managemen, because it's just a placebo? I've always considered that if someone has a pain that can be eased by the use of panadol then they never really had a pain. Honestly you might as well have a jelly bean. Or is it just me who is totally resistant to it?

Several studies involving double blind testing have proven that paracetamol is more effective at reducing pain than a placebo. Your resistance could be due to a variety of factors, maybe you should take a higher dose, or maybe you have a unique version of the COX gene that isn't really affected by paracetamol. Try taking a different analgesic like aspirin, ibuprofen or codeine.

I do find limited effectiveness with aspirin, however as it's not recommended to be taken by gout sufferers, which alas I am, I steer clear of it . Ibuprofen works fairly well and voltaren is quite effective. Codeine was always a safe bet but now, of course, it's not readily available. I pretty much only use pain killers for the odd case of gout or bursitis. Thank you for your reply.

Interesting and possible, however I have recently found out from speaking with chemists that I am not in the minority. Apparently many people don't get any relief from paracetamol. This has been highlighted by the recent restrictions applied to the sale of codeine in Australia. I'm not of Asian descent. Thanks for your input.

Dusk Some Asian people don't lack highly important digestive enzymes, they might just work a little differently
