How do you know it's a first print run?

The seller I bought it from had the case the booster box came in. The case tells you the print information. With just the booster box alone, I do not know how to identify it's a first print run. There supposedly is a way to tell, but it seems to be a tightly kept secret and it's perhaps the number one question I also have remaining in my quest for answers in vintage collecting. I've dealt with several boxes now that are confirmed (by case) first print run and I can tell you I don't see a difference between those boxes and other boxes that were of unknown print run. If you figure it out, let me know!
