Love the blaagar, i have a few odd recipes i refer to as slam tech. Lol pretty much 2-3% of watever i have laying around. Have even used the water from boiling black beans, also love using the run off when i ferment wood chips for my outdoor beds. For real anything that is carb/sugar/starch/yeast based will work

At 20:00 your adding lots of nutrients. This may have an adverse effect on your growth due to the ability of bad bacteria or fungus to thrive eating those same nutrients. Sometimes less is more. It's usually a good precedent to test this out in smaller batches and get your choice of culture stable prior to only using this recipe. Always have a backup plate ready in case things go wrong. Good luck everyone and learn before you burn. Mycology is awesome 👍😎

Thanks for the video it was very informative, I especially appreciated the detail on condensation, I noticed the scalpel blade was burned at the tip but no carbon deposits, I use a candle to sterilise but always have a build up of carbon. It looks really messy, especially with spores. Any tips for getting the blade sterile yet ‘clean’, thanks

Fascinating. When I first got in to growing mushrooms I didn't realize that I'd have to become a micro guy! I just started, I injected spores into my substrate x4 and nothing happened. Great first failure LOL.

So, I never have an issue with the media bottle. I always had issues with pouring from a jar. Now, i have to sut my media bottle at a tilt in my PC. I use a small jar to keep it lifted. While also keeping the lid slightly loose with foil on. Never had it boil over.

imho and maybe and all due respect and thank you for the video The video shows the petri dishes held upside down. The larger dish should be lower and the agar should coat the smaller dish the way that the ceiling of the smaller dish is coated. This way microbes cannot fall on the surface of the agar to avoid contamination. This is routinely done in microbiology laboratories. Petri dishes with microbiological medias are routinely stored like this before inoculation during storage of sterile condition, and after inoculation during incubation. Mycological specimen are usually incubated on room temperature (18 -20 dec C) bacterial media is incubated at human body temperature (38-40 dec C). At 38 deg C gelatin melts, agar-agar stays solid. Gelatin media is probably useful for mycology specimen, because it stays solid at 20 deg C incubation. Obtaining gelatin, jello, is easier in an everyday setting, and it is useful for incubation on petri dishes. It may be a kind of biiohack.

Yes. You can but keep in mind each exposure to open air will possibly introduce contamination. It also may be a bit clumpy depending on your recipe and age of agar.

I use grafting tape instead of the specialized tape. I guess it's only an issue if you do no-pour agar?

As a retired lab-rat turned mycologist, I've always used parafilm until recently. I switched to grafting tape and find it is much cheaper (0.4 cents per plate vs. 10 cents per plate for parafilm) and much faster since I don't have to cut the grafting tape into strips like I did for parafilm. Takes a little getting used to if you're already familiar with parafilm but well worth learning. Since grafting tape is so cheap I do seal every plate after the agar is set before storing. Something I didn't do with parafilm.

Oh man, yes, you're correct. That's an embarrassing and misleading mistake. 0.01%-0.05% is the correct dilution, 0.3% is way too much! I can't believe I didn't catch that. I'll post a correction in the description, thank you.

do you have any tips for eliminating condensation when you only have a SAB to work with? pouring at 115 helped a lot but I still get some moisture and then I end up seeing contam where that moisture is sitting.

Thank you for the video. I think, regular jello should work too. Regular gelatin melts at 38 deg C in an incubator. That temperature is used for bacterial growth copying human body temperature. Fungal cultures are incubated, usually, at room temperature 18 deg C to 20 dec C. Gelatin stays jello.

I watch these videos but remember when I watched someone just take mushrooms and chuck them into a blender and layer them with some media in a pattern and then watered them to end up with mushrooms in a few weeks. How is the lab method different or superior? Just wondering if it does anything special.

You have some very beautiful mycelium agar plates my agar plates always get thick fluffy growth with long thin strands stacked on top of each other yours has a nice thick ropey growth that's very beautiful does this mean the cultures are strong and healthy or does it mean they are hungry because I've followed some of your recipes and its given me very happy looking fast growing thick and fluffy but not as beautiful as your plates how do you achieve that and is that a good thing ? I never knew that about the hydrogen peroxide I am going to give that a try I still get contamination from time to time maybe 1 in every 10 plates is still becoming contaminated with trichoderma. Could I spray say 0.5 -1% around an attempted clone that became very contaminated and grew some very scary looking stuff but also grew a spot of mycelium if I cover it spray with 1% around it then in 24hours spray 0.3% directly on the mycelium before doing a transfer to clean agar with a very low sugar content and 0.3% H2O2 will it grow without contamination spreading with it and if the contamination is all ready embedded will it atleast slow the growth of contamination and allow clean mycelium to grow ? People say just do water agar transfers but if mycelium is Gona grow on it so will other contaminants and they can in alot of cases out grow the mycelium so would you personally consider this the go to method over water agar and would you recommend anything else to help slow contamination but aid in mycelium grow eg pine contains a natural disinfectant garlic has antibacterial properties cinnamon is a mild anti fungal and ginger is natural disinfectant with 1000s more do you know of anything like this ? Does sprinkling a bit of saw dust and small grains on the agar plates before sending to colonize grain help at all? Is there anything other way to dry up the agar plates from the condensation that's not sitting Infront of a flow hood I've not got 1 and use a still air box for transfers so what's a good way to remove the condensation from my petri dishes before I transfer anything to them ?

imho and maybe and all due respect and thank you for the video Isolation of microbes: a mixed culture can be inoculated on the surface of the media about 1/3, 1/4 of . After that the inoculation loop is sterilized and from the inoculated surface using one streak the microbes can be carried to other, sterile side of the media surface in the petri dish. This produces a dilutional effect and the microbial growth shows up on the diluted surfaces a single colonies making it possible to transfer colonies of a single organism to another petri dish. Mixture of microbes can be isolated into colonies grown of single, one kind of microbes. Pure colonies can be obtained this way. imho and maybe and all due respect and thank you for the video

33:45 Why are you discarding agar EVER? If you are willing to put Dung and Dog Food in your agar, isn't fair to say "anything goes"? I've started saving ALL of my waste agar, ALL of my contamination, and recycling it. I add agar or water to adjust the consistency as needed. AFTER cooking it at 20PSI I compare the taste to pure 2% LME water and pure 2% Honey water to estimate how much sugar it has and if any needs to be added. I'm getting LESS contamination with my "Waste Blagar" than I did when I was trying to be perfect. I'm getting so little contamination that I have nothing to add to each cycle of the agar. I have started breeding contamination in jars just so I have enough to add to my Blagar.

You're joking. Your comment is intentionally misleading. (The gelling agent in the agar is a polysaccharide, or two polysaccharides, formed from a sugar, (galactose?) linked in long chains, I think? You have some knowledge of biology.) (Agar may be found in oriental food stores, or certain health food shops. It is only part of the culture medium mixture.)
