When mapping controls say R3 button to back paddle does the R3 button become blank?

No, it still works. But the back buttons mapped to the R3 can only press.

After you cancel the default remap, how do you apply it?

Re-enter the setting mode, and set up the remap layout again.

Can more than one button function be remapped to one paddle? (i.e edit and reset from 2 separate button functions be remapped for a single paddle requiring multiple paddle presses to complete the second function remapped)

No, you can only register one button function to one paddle at one time.
