For future reference: Alcohol will sap moisture from the specimen. It is fairly simple to re-hydrate a specimen by placing it in a sealed container with moist paper towels. Just be sure to keep the specimen from directly coming in contact with them. 24-48 hours later and the insect will be easy to pose and all the joints should be flexible. If you are close to home and not on holiday, placing your specimen in the freezer is an effective way of dispatching it.

I just finished looking at the mosquitoes with a microscope, and it felt terrible. Especially the fine hairs on the mosquitoes, it feels particularly uncomfortable! It seems to be a monster in some science fiction movies, it's terrible!

HINT: before embedding use KOH or NaOH for claearing the keratinscelet and dont use the care Eupral. Make your own glyceringelatine with small amount of sorbic acid for conservation

As you mention, easy to inadvertently lose parts, so I found it useful to wear a readily available face mask when I dissected a mayfly recently.

Hello. Yes, I have used the concave sides, but are only good for very small insects. Sandwiching two coverglasses with a spacer between is by far a better method. Fixing the insect between them with mountant preservative allows us to view the insect on both sides with equal clarity. Concave slides are viewable only from the top side through the coverglass. My technique is the winner, me thinks.

To expand a little on my previous comment, the DPX smells just like the resin we use to do minor car-body repairs. I gave it around 48 hrs to set before touching the insect specimen. The DPX had become rock hard by then and like glass.

I would assume you would keep these upside down while they dry so the parts would be up against the cover slips, right?

If you use little mounting medium, then they will touch the cover slip anyway. Gravity also helps.

Hello, again, Microbe Hunter. My original post seems to have gotten lost. KZbin confuses me sometimes. Anyway, to my question: you said that to use DPX properly with insects I would need to allow the insect to dry out, then teat it with alcohol and then with isopropyl alcohol before finally using the DPX on it. Can you tell me the reason for all these processes?

The alcohol treatment is to remove water in fresh specimens. If the insect is already dry, then this is not needed but can not hurt because the insect might be less brittle. The isopropyl alcohol (isopropanol) is needed to completely remove water because the previously used ethyl alcohol (ethanol) still contains a small percentage of water (isopropanol is more "dry" than ethyl alcohol). Before DPX it should be placed in xylene for some time, or if this is not possible, soak it directly in DPX before mounting. This ensures that the DPX reaches all parts of the insect. The problem is, if moist specimens are mounted in resin media (DPX) because then the water in the insect can not evaporate and bacteria might decompose it and this will form gas bubbles (decomposition gasses) that push off the cover glass again. The alcohol therefore not only removes water but also kills off bacteria in those parts of the insect that might not come in contact with the DPX.

Not here, but parasites are nothing unusual. Mites are a real problem for bee colonies etc.

@@Microbehunter: I have got photographs of said mites, five of them inside the head of a Gall Midge, of the family Cecidomyiidae, itself no more than 2mm long. The parasites look very similar to ticks, but at a microscopic level.

I used Eukitt, but one has to be careful because of the fumes (fresh air needed). Euparal is more commonly used to mount insects, however. I wanted to try out Eukitt this time.

Yes, I know. I tried to suppress the room echo with the audio editor, with limited success. Today I found a setting on the camera that improves it somewhat (wind suppression).

@@Microbehunter: Some folk just like to criticise. It doesn't sound horrific at all. Nothing wrong with the sound. It was clear enough and absolutely fine to actually talk about and hear the subject we are interested in. It's not a Hollyood movie we expect to see or hear.
