The guide you found steered you wrong, my dude. You can absolutely go back into the level, but you'll have to finish it again to collect that last pill, and it will require the coin. There is a line of carts near where you first enter the store that takes a coin to unlock, and one of them will have that last pill. As a general rule, it takes anywhere from 10-20 hours to complete in it's entirety.
