Drugs Shown to Inhibit SARS-CoV-2 in COVID-19 Disease: Comparative Basic and Clinical Pharmacology of Molnupiravir and Ivermectin
austinpublishinggroup.com/pha...
/ leslie-adesuyi-ajayi-m...
There are still many nations where vaccines are not yet widely available,
There is a gradual shift in focus, to antiviral drugs,
Adjunctive chemoprophylaxis
Active treatment of new SARS-CoV-2 infections
Post -vaccination breakthrough COVID-19 cases
The two ways to get new drugs
Develop novel antiviral drugs for SARS-CoV-2
Repurpose existing FDA -approved drugs to treat COVID-19
Ivermectin is the most studied “repurposed” medication globally,
in randomized clinical trials, retrospective studies and meta- analyses.
Molnupiravir and Ivermectin Anti-SARS- CoV-2 Mechanisms, Pharmacokinetics and Pharmacodynamics
Molnupiravir is a broad spectrum antiviral agent against SARS- CoV-2, SARS-CoV,
seasonal or pandemic influenza and MERS corona virus
Ivermectin is an FDA-approved, WHO essential drug used as broad spectrum antiparasitic, antibiotic
and which has demonstrated broad spectrum antiviral activity against RNA viruses, including HIV, Zika, MERS corona virus
The FDA-approved drug ivermectin inhibits the replication of SARS-CoV-2 in vitro
www.sciencedirect.com/science...
5000-fold inhibition of SARS-CoV-2, (99.98% at 48 hours
The inhibitory concentration IC50 of Molnupiravir shows it to be a more potent anti-SARS-CoV-2 agent, compared to Ivermectin in vitro.
Both molnupiravir and ivermectin are well absorbed after oral dosing
Tmax of molnupiravir being 1-1.75 hours,
With a half life of 7 hours
Tmax of ivermectin is 4-6 hours
Very long half life of 81-91 hours
Ivermectin, being lipophilic has a large volume of distribution
Ivermectin has the ability to accumulate in the lungs
The anti-SARS-CoV-2 actions, both of molnupiravir and ivermectin, are dose and concentration dependent
Molnupiravir active metabolite (NHC-5’ Triphosphate), acts as a competitive alternative substrate for viral RNA
causing viral mutagenesis or mutations, which leads to viral error catastrophe and extinction of replication
There is some concern about the safety of NHC -nucleoside triphosphate, which is also mutagenic to mammalian cells
Ivermectin, multifarious actions,
Binding to SARS-CoV-2 spike protein S
Reducing cell entry via human ACE2 receptors
Reducing viral transcription
Inhibition of cytokine production and inflammation
(not yet been shown for molnupiravir)
Complimentary pharmacokinetics and pharmacodynamics of the drugs
May be additive or synergistic
This should be further investigated in anti-SARS- CoV-2 antiviral combination therapy.
A combination of molnupiravir with Ivermectin putatively, in effects on RdRP or cytokine release.
Cost

@@ProfSimonHolland One of the reasons we have had trouble seeing efficacy when “studying” ivermectin against COVID-19 is dosing is too low and duration of dosing is usually too short in all studies, and the patient is not followed long enough. The course should be a minimum of five days, and the dosing should be higher than what is used in parasitic treatment, body weight dependent. Ivermectin’s efficacy becomes obvious on day six of use. Recent study from Israel followed patients for 15 days on only a three day dosing showed remarkable efficacy versus placebo. On an anecdotal note, I saw this timing and advocacy of ivermectin when my wife was treated for COVID-19 . Her doctor prescribed ivermectin, she was on it for 10 days, day six and beyond was key.

When a Japanese medical group offered to collaborate with Merk, (the original inventor of Ivermectin, however who's patent has snice long expired) and to do trials and studies on effectiveness of Ivermectin against COVID-19, Merk flatly declined. Now here we see Merk have developed a similar drug they are pushing to get approved and at nearly 1400 times the cost? This speak volumes about the integrity of these pharma groups, and the very Governments that are willing support such behaviour.

Within hours, our major symptoms subsided, less the back pain associated with the delta variant. We followed up with a second dose after 48 hours and I am completely asymptomatic and my spouse, who was bedridden prior to administration of IVM, has recovered to the point of mild symptoms, with some fatigue. We had asked our primary to prescribe IVM, and we were met with denial because “there is no scientific evidence that IVM has any effect on the virus.” The fact that this existing drug is being denied to people who are ill with COVID-19 is criminal.

In all fairness, the push for mandatory vaccinations and Pfizer's attempt to get FDA approval to vaccinate children as young as 2 years old makes it difficult to give them credit for having any moral integrity whatsoever. The behavior of the people pushing vaccines doesn't convey any resemblance to scientific scrutiny or medical responsibility. It is worth doing a study to try to determine if everyone would have been and would be better off without any vaccinations at all, while relying on natural immunity and ivermectin, which is what 3rd world countries have been doing anyway. Obesity is a subject that hasn't gotten enough attention as well.

@@taitjones6310 I'm sure comparisons will be done between nations with the different approaches. Of course, there won't be any pharmaceutical companies paying for those studies unless they're quite positive the results will favour them. But there has been tremendous scientific scrutiny on the vaccines. Moreover they are one of the few things which are incontrovertibly proven to help the situation. In my little home province we had a nasty run with Covid earlier in the year and the vaccines we purchased were temporarily delayed in delivery. So when they did become available the majority of people got the jab. The statistics trend rather clearly. On the one hand, vaccinated people make up 40% of cases in our province. But 80% of the population is vaccinated, meaning 20% of the population comprises 60% of the cases. When you look at critical illnesses the differences are much more pronounced. We rarely have anyone in our ICU's who is fully vaccinated. We also notice that areas in the province with lower vaccination rates have higher case rates. As someone who is scientifically inclined, I have no grudge whatsoever if Ivermectin or any other treatment turns out to be more effective than the vaccines. But we do know with certainty that the vaccines help.
@taitjones6310
@taitjones6310 2 жыл бұрын
@@trentreimer130 There are A LOT of different things that factor in to a person getting infected, but I'll try not to run off on a tangent on this subject. As far as vaccines are concerned, in the Pfizer clinic trials that I read the results of, the vaccinated group had 5% fewer infections than the control group. This is where the "95% efficacy" claim comes from. I know it's misleading and counterintuitive, but that's the difference between vaccinated and unvaccinated (at least in the published clinical trials). The point being that the media has facilitated recommendations from Big Pharma companies via the WHO and CDC, who have been adamant about telling people that common sense and time tested things like taking care of your immune system through exercise and fresh air along with vitamin supplements of D and Zinc are "pseudo-science". As well as demonizing drugs like ivermectin and hydroxychloroquine. Just because it needs to be said, I'm not in the boat of believing that the 'vaccine' is a cloak and dagger means of mass murder, but it is a cash grab. There hasn't been any scientific scrutiny publicly over the difference (long-term) of just letting everyone get it while finding the most effective methods of treating the infected. Most notably, there is a false idea that anybody, vaccinated or not, is going to escape getting infected. EVERYONE WILL GET INFECTED. It's an inevitable fact. By this point everyone has already been exposed to it, and it's going to be around as long as humans are, so our bodies have to adapt to it, and that is done through exposure. To summarize: drug companies are focused on selling as many vaccines as possible to pad their profit margins, and the argument over vaccine efficacy is a red herring. The issue everyone should be focused on is supporting and improving their immune systems and overall health, and I haven't witnessed much scientific scrutiny over that.

Governments around the world have one purpose: to sell gratuities to the highest bidder. At last, the people are on their own! Medicine in the USA is more corrupt than Wall Street Clearly the breadth of Wall Street fraud is extraordinary. I submit medical fraud is equal in breadth but more pernicious in scope. This is because everyone signs onto the gravy train; and, it feeds like a parasite directly off the misery of the people. “Come in here, dear boy, have a cigar You're gonna go far, you're gonna fly high You're never gonna die And did we tell you the name of the game, boy? We call it Riding the Gravy Train” Why doesn’t a dentist treat the infective agents that ‘cause’ gum disease with the required antibiotic combinations - or any antibiotic? Why are physicians’ access to medications and antibiotics (e.g., amphenicols) blocked? Why are vitamins like E fraudulently demonized whilst drugs that shut down hormone production (statins) - resulting in premature aging - approved? The number one cause of emergency room visits - blood thinners - shouldn’t even be legal; there are in fact, far safer solutions! Cures are blocked; and, treating the underlying ‘cause’ of disease is blocked. The profit is in the management of symptoms at best; and, creating more disease at worst. In fact, there are countless examples of surgeries and drugs that are deployed to temporarily mollify symptom; but, which ‘cause’ far more long-range disease and suffering. All of this is great for profits! No one questions anything. I wrote a medical text about psychotropic drugs. In my research I documented 100s of autopsies that gave evidence that benzodiazepines were the seminal cause of death to many celebrities over time: Prince, Elvis Presly, Michael Jackson, etc. Benzodiazepines form a chemical torniquet around clusters of neurons in the brain. Over the short term this can have the effect of lowering anxiety as brain transmission is greatly attenuated. However, over time, this kills enormous patches of brain matter. As large patches of the brain die the patients mind grows weaker; and, they must resort to opioids and/or alcohol to face the challenges of daily life. Brain death in long-term benzodiazepine use has been clearly elucidated and shockingly visualized thanks to advances in brain imaging studies. After taking benzodiazepines for a time, the sleep architecture of the brain fails requiring the patient to take yet more brain-damaging drugs. Death by suicide or overdose is typical. For this reason, the European Medical Associations have blocked the long-term use of these drugs. Yet in the USA, Xanax is a money maker: priced at over 30,000% higher than the international generic equivalent. No one questions when doctors randomly remove masses of organ tissue; or cut up vertebra resulting in their collapse around exiting nerve bundles. Not only did their procedures fail to consider & address the underlying ‘cause’ of disease, these procedures effectively created a highly-profitable life-long in-patient phenomenon. Government supports it all because big pharma monopolies, medical consortiums and bankers who back it all fund elections. Today we shuffle barefoot through TSA lines whilst trying to hold up our pants; and, socially distance, wear masks that damage viral fighting immunity, and close businesses simply because government bureaucrats tell us too. No one is permitted to question anything. In many cases, if you want to ‘get well’ or achieve good health you have to leave the USA for treatment. The bureaucrats block treatments. Take the example of Lyme Disease - which was in fact created by the US Government: the cure is banned by the IDSA, (Infectious Diseases Society of America). Chronic Lyme patients typically suffer from multiple infections. Therefore, the cure to chronic Lyme disease requires either an amphenicol or multiple antibiotic drug combinations to cover the breadth of coinfected agents, (e.g., Anaplasma, Babesiosis, Mycoplasmas, Bartonella, Tularemia, etc.). However, the IDSA will fine a physician $10K and revoke his license unless he treats the patient with (only) doxycycline. And yet doxycycline does nothing to kill the many co-infective agents or even borrelia (borrelia merely rolls up into a protective cyst until after the patient discontinues taking doxycycline - then it comes back out to infect the patient). The Connecticut AG sued the IDSA for this practice - finding five executives there to be holding corrupting financial interests in the big pharma monopolies. And yet nothing has changed in this policy set by the IDSA: physicians are still banned from curing the disease. There are countless examples that can be cited. The Americans pay a lot, lot more simply to be parasitically sucked dry by an inhuman system. If you want to get a cure, you have to leave the country! It was for this reason that I voted with my feet, and moved out of the USA for good - Which has come to be the best decision of my life. The U.S. Is an “Oligarchy With Unlimited Political Bribery” Former president Jimmy Carter said on the nationally syndicated radio show the Thom Hartmann Program that the United States is now an “oligarchy” in which “unlimited political bribery” has created “a complete subversion of our political system as a payoff to major contributors.” Both Democrats and Republicans, Carter said, “look upon this unlimited money as a great benefit to themselves.” Carter was responding to a question from Hartmann about recent Supreme Court decisions on campaign financing like Citizens United. Transcript: firstlook.org/theintercept/2015/07/30/jimmy-carter-u-s-oligarchy-unlimited-political-bribery/ MEANWHILE: Vaccine monopolies make cost of vaccinating the world against COVID at least 5 times more expensive than it should cost! Find me a doctor that has treated 400+ patients with Ivermectin and believes it doesn't work. It's definitely not a 100% miracle cure all drug. @@MikeSmith-vy4wp Most doctors prescribing Ivermectin are prescribing it in addition to other treatments, making it impossible to tell if it was the Ivermectin that actually made the difference. Additionally, not all patients who are infected with COVID go on to develop severe disease so looking at any one individual who takes Ivermectin actually can't tell us anything about whether it actually works. They might have just had a case that would resolve itself on its own without intervention. But really what you are talking about is the individual call of a doctor after weighing the specific risks and benefits of the drug to his or her patient.
