Very good explanation. But, does the Lipinski's rules of 5 not include the polar surface area of a molecule?

Oh no, it doesn't. That's Veber's rule.

But I read that the 5th one is : molar refractivity should be between 40-130

There are a bunch of different varieties, but the key take-home message is that Lipinski hasn't led to a new drug in ages.
