What is being extracted? I thought the sodium bicarbonate was being used to do the extraction from the methylene chloride...why does the methylene chloride need to be evaporated at the end?

hey man! Love the video and the way you present it. I have a problem though... I'm doing this liquid liquid extraction of a carboxy benzo crown ether out of a ethanol-water solution after a reflux. The extraction is done with chloroform. The problem is, normally the carboxy benzo compound should be in the chloroform phase after extraction and evaporating the chloroform doesn't allow my crystals to form. Protocol used is this one: ''Cation Binding Properties of Crown Ether-Modified Polyet hylenimines'' on page 856. Any suggestions on how to get my crystals? This is for my thesis paper in University and time is ticking! Thanks so much

I find you tube an excellent resource when preparing for chemistry undergraduate practicals. One is able to obtain a feel for the techniques when they author explains in detail their actions and techniques. In my limited experience as a chemist I've found that little adjustments to the way procedures are carried out cumulatively make a big difference. Have you done a flash column chromatography one?and nitrophenylate ester hydrolysis?

unlikely, this extraction is good for solution. However, we know that Westphal et al. applies this exact same liq-liq extraction using phenol-hot water to extract endotoxins from bacteria culture.

Lectin is a big protein, I presume it might precipitate in the solvent, possibly other methods could work.

+Genar Gundan This is an extraction protocol, we can grind the dry product using mortar and pestle, and use an organic solvent to dissolve the extract, after which, conduct the extraction procedures as shown in the video above.

Hey so I have an unknown compound. I want to do an acid base extraction on it. I think the compound is basic because of the solubility tests I did. Do I use HCl instead of NaHCO3 when adding to separatory funnel? thank you

+Samuel Lee if the organic compound you wish to extract is basic, first, add HCl to form a salt, so it goes into the aqueous phase. we get rid of the organic phase to eliminate organic impurities/ side-products. then, take the aqueous phase and add NaHCO3 to convert your desired compound back in to the neutral form (no charge) so that it goes into the organic phase, then collect the organic phase (so your aqueous impurities remains in the aq phase) --> rotovap solvent and TADA! see kzbin.info/www/bejne/foS9e5asbcuaa8U

@@mantiger11 this is late, but you could vacuum distill the solvent if you don't have a rotovap or with low boiling solvents you could also just use atmospheric distillation

Hi Fun, I'm doing research on extracting peptides from fish fillet for antimicrobial study, one of the procedure require me to do acidic extraction,which I need to remove acid from the organic solvent,can I use this method? What's your opinion?

Sounds ok to me, give it a try. Acid remove the amines in the fish. ensure that the temperature is not too high, if not it might cleave the peptide bonds.

Sir i have a question, When can we say that the lower surface is Organic layer? because the Water Density is higher than organic solvent. Because some reference said that Organic layer is the upper part. Thank you

use density. denser layer will be at the bottom. most organic solvents are less dense than water. a few, e.g. dichloromethane, trichloromethane are denser. so be clear!

When I try, it allways clump. Even when snowstorm effect happens, some parte of magnesium sulfate clump. Why? Is it normal or it does means that drying wasn't efective? Thanks if someboddy help me

If they clumped together, this is an irreversible process. no matter how much more you are going to add, these remain as clumps.

I have a question since your clearly a professor. what would you recommend for safety precaution's for various alkane nitriles such as acetonitrile etc. I know of course a good fume hood but with a good fume hood would doing reactions with such alkane nitriles be enough to prevent the worse case scenario (cyanide poisoning) What else would you recommend for safety. Any tips or what you would do would be great!

Danny Esliger Hi Danny, I am extremely regretful I just saw your reply, I hope this will still serve you good.The list is quite long (safety precaution for the chemicals aforementioned). What we normally do is to find the MSDS from the internet (many chemicals companies offer updated details) and sieve out those safety information that are pertinent to your students/ projects/ experiments. cyanide poisoning would be avoided as this is a controlled item. : safety data sheet :

how is the organic layer in the bottom and aqueous in the top ?! since, organic layer is the lighter ! it should be the upper layer !

The organic layer for this video is Dichloromethane (specific gravity = 1.3266 g/cm3) vs water (1.00g/cm3) so the organic layer is denser and stay at the bottom. In other extraction, using solvents such as diethyl ether (0.7314g/cm3), the organic would be at the top. We should always check the density before doing extractions to prevent losses.

Tell that to the 5 technicians that I worked along in my career and their families. They all had some form of cancer and died FROM working with chlorinated solvents, one GC- MS Chemist died from brain cancer . Put on the viton gloves or don't do the organic extractions.
