In this video I had the absolute pleasure to chat with Professor Valter Longo, Director of the Longevity Institute at the University of Southern California -Leonard Davis School of Gerontology, Los Angeles, one of the leading centres for research on aging and age-related disease and the Director of the Longevity and Cancer Program at the IFOM Institute of Molecular Oncology in Milan, Italy and also author of the popular book "The Longevity Diet".

@pr k "Zoledronate is a nitrogen containing bisphosphonate, which inhibits the farnesyl pyrophosphate synthase (FPPS) enzyme, central to the mevalonate pathway. It is already used clinically to prevent fractures in osteoporotic patients, who have been reported to derive unexpected and unexplained survival benefits. Using Drosophila as a model we determined the effects of Zoledronate on lifespan, parameters of healthspan (climbing ability and intestinal dysplasia) and the ability to confer resistance to oxidative stress using a combination of genetically manipulated Drosophila strains and Western blotting. Our study shows that Zoledronate extended lifespan, improved climbing activity and reduced intestinal epithelial dysplasia and permeability with age. Mechanistic studies showed that Zoledronate conferred resistance to oxidative stress and reduced accumulation of X-ray-induced DNA damage via inhibition of FPPS. Moreover, Zoledronate was associated with inhibition of pAKT in the mTOR pathway downstream of the mevalonate pathway and required dFOXO for its action, both molecules associated with increased longevity. Taken together, our work indicates that Zoledronate, a drug already widely used to prevent osteoporosis and dosed only once a year, modulates important mechanisms of ageing. Its repurposing holds great promise as a treatment to improve healthspan." This is from "Next big future" source from "Lifespan.io" This can possibly increase lifespan by 5% in males, and 16% in females, and only needs to be administered twice a year.

Bisphosphonates do extend life if taken in middle age and according to the research on fruit flies, it would benefit women much more than men. We need more research on this.

You don't have a lot of subs, but you've got some excellent knowledge and enthusiasm. Would echo David Liu's thoughts on you at the end of his interview - you're very good indeed. There isn't a lot of good content on your space on youtube.One thought would possibly to do something on the other elements of ageing - outside of the NMN, mitochondria stuff - i.e. sun protection, damage and cancers from the sun, tretinoin and associated vitamin a derivatives, c+e serum; just to bring in some more subs i.e. if they're searching for "how to get rid of wrinkles", you can then bait them into your broader content.

I agree with Dr. Longo, I am from the Mediterranean and I grew up eating lots of vegetables, nuts and fruits, whole grain corn bread, yoghurt and occasionally fish, of course with olive oil.

Seeing how much you are already involved in the area, makes me look at you like you are already living in an "Altered Carbon" type of Era, while we struggle to keep up. To comment on the bone strength study... it's called "Vegetarian and vegan diets and risks of total and site-specific fractures: results from the prospective EPIC-Oxford study" Pescetarians are shown to have slightly better results than "meat-eaters". What is forgotten to be mentioned is that like always "meat-eaters" simply means "all the remaining people" after selecting out groups of vegans, vegetarians and pescetarians, who make a conscious nutrition decision. The rest does not. Which means there's a very severe "healthy user bias" against "meat-eating" group. If you would select out of those health-conscious meat-eaters, equivalent to other three groups, they would most likely easily dominate the results. Then a completely different topic. I can hear you now have a very solid quality audio, wasn't always like this. What microphone model is this?

Mark Mattson. My favourite longevity researcher. He is not selling ANYTHING, not even books he has authored. He is a public servant and said his work should be freely available. No profit motive at all. I recommend you check him out.

Notice how he is associating low protein to activity which is indirectly associated with calorie volume.basically I f you are active then you need to consume more protein ..excess protein is converted by the body into a fat not into insulin ..that’s why most people whom incorporate resistant training during fasting periods (like myself) can still enter ketosis even though calories are being consumed

@@scotlandtheinsane3359 I think that is a very narrow conclusion based on 1 population instead of taking in account the entirety of the Blue zones like Dr. Valter Longo just explained. You cannot take just 1 and conclude it works like that. Also Hong Kong has the highest Female life expectancy to be correct, for males it would be Switserland. Also this is only taking account Life Expectancy as an average. But "longest lived populations" are not only determined by Lifespan but also healthspan as well as having a longer livespan over the history of the population. So not only now as an average but having a long history of said average in that period. Lastly.. If you take the averages of live expectancy of those specific regions they study the average lif expectancy has been that high for decades now, not just in increase over the years. Also do not forget the native cuisine of Hongkong has a huge plant based component where they are actually expecting live expectancy to drop again when the older population starts to disappear. If you take the health statistics between the "young generations" and compare that to the same period from the now old populations you get a staggering difference in disease occurance. So no "They eat alot of meat and everything" is actually showing more disease in younger populations within Hongkond.

@@scotlandtheinsane3359 "the end" wow you really lack knowledge about how groups work within epidemiology and cover it up with being smug and overconclusive. Even though in science we never make straight 100% conclusive claims. I am going on a limb here and explain. Let us say we got our 'old' population they are all 70+ right now and grew up predominantly plant based like Asia was throughout the 1950 - 2000's. These people get old and do very well of healthspan. Now the second group 40-70 they were raised on a plant based diet but moved to this 'new' diet over time. They are doing well but not as well as th 70+ generation. Finally the 'young' populations 20 - 40 had a plant based youth but moved to the new diet much sooner in life and they have the western disease occurance as in Europe. Now when we view them seperate we see age related disease progressing faster and faster where the groups entered the new diet sooner. And now the crux. When we COMBINE the results the old compensate for the young groups, giving a high average. This is the Simpson paradox where one grouos aspects disappear when you bunch everything up. Problem? The average is not representative of the new diet and will decline once the old group starts to fade and stop compensation. The issue is that they tend to live so long they skew the results for a long while. Expectetations? They healthspan and expected age will drop massively in populaion in Asia that were plant based long time before the wester diey hit them. Hope you leaned something instead of reading with you bias.

I've listened to several interviews that Dr Longo has given and as a student of Dr Roy Waldorf's I respect his opinion. As a regular water faster I've often wondered why he would choose foods that where not more towards the ketogenic spectrum. Of course I could be wrong but I thought ketosis was a marker of autophagy? It makes sense that if FMD is a product that you're marketing to the masses that you'd choose foods that appeal more to the most people. Excellent questions! This interview cleared up a lot of questions I had about the FMD. Thanks!

Notice his opinion of veganism, which he describes as difficult to do properly. I guess he applies this to keto/fasting as well. And his stance in general seems to be "doable by average people," as opposed to optimal. In other words, i think only recommends super easy diets... I say go veg, take your B12 and D, eat low carb, and fast 36 hours 1x/week!

@@DIYDSP unfortunately we're well passed the bartering system so I can understand his motivation. In the end I'm going to do what I believe is right. My concerns are health span and sustainability. Really I just don't want to die a horrifying death. Be it shot to death, living out my years in some type of prison (in all their forms) or being run over by a combine! Lol.

Keto diets are too high in protein. For the FMD he wants to go very low protein for the five days, which mimics water fasting.

@@stargazerbird then why not stick to more fat... High protein causes glycogenesis. A true ketogenic diet is low in protein by anybody's standard. I'm not try to be disrespectful, just trying to understand. Once again, this video cleared up some questions I had. Research ain't cheap!!!🤓

Strangely two vegans I know have both ended up with cancer. He’s in remission from testicular cancer and she’s been given less than six months. Brain tumor stage four out of nowhere

That depends on the fish you're eating. Sardines Scallops Squid Salmon Oysters Salmon Trout Sole Flonder Clams Anchovies Are not much of a concern in regards to heavy metals.

You mean having like 175 grams of protien in one sitting? Assuming that’s what I need yo bulk up and build muscle? Is there anything showing this works? I’ve been trying to make sense how to balance wanting to build with giving mtor a rest now and then.

Where can I read about the "negatives" on 16H fasting? It is really hard to find anything on the bad sides of 16H fast! And it seems there are not a lot of Benefits at all to a 12H fast?

Yeah I’m doing 22:2. Valter said lots of bad info about 16hr+ fasting - I was like wtf? I wish he said what was wrong with doing consistent 16+ hours fasts.

I understand the F.M.D. is a compromise to maximise adherence whilst training some benefits of fasting e.g. the calories and sugars, but I wonder if adherence wasn't an issue, what would the optimal F.M.D. be? Lower carb? Lower calorie?

The optimal is water fasting. The FMD is designed to be very low protein, high fibre and moderate sugars and high healthy fats but also very low calorie. This means you can achieve deep autophagy which gives you the same results as true water fasting.

Id say no. Intermittent fasting can have benefits but if you are already healthy then there isnt much need. Currently i am doing a 48 hour fast which im doing only because i was having problems with my gut and need to "reset" it. Im trying to increase my HGH and also reset my stomach receptors because no matter how much i ate i was still hungry. So im doing this 48 hour fast for the first time which is something everyone should do once a month or so. So far it feels like im feeling better and i definitely needed this fast because my body was really out of balance with my hormones

If you keep continuing to lose weight in perpetuity, at some point, you will be dramatically under weight. You want to avoid that. The medical establishment would probably say that your IF practice is okay as long you stay within the healthy reference ranges for your biomarkers and weight (bmi). If your biomarkers are starting to fall out of reference ranges due to undernourishment, then a problem would exist. You definitely want enough nutrients to keep bone mass. My understanding is that bone loss is irreversible.

If you eat crap then yes. I fast 18 hours and eat two nutrient dense meals.

Good interview but I find Prof Longo gives conflicting messages - an example being if you fast for 16 hours. I fast between 18 & 23 hrs daily & I know the feeling of ketosis & know that autophagy is doing its thing. I heard him in other interviews saying 12 hrs is better than 18, 24 or 36 hrs! I think he suggests that the negatives lessen once you get to 2-3 days & deeper into autophagy. I'm no expert (of course!) but isn't it unfair to demonise high protein or high fat? As far as I'm aware, most studies showing poorer outcomes when animal protein is high doesn't prove much to me because I bet virtually all those subjects also ate high carb, drank alcohol & most likely, ate junk & processed foods too. If done correctly (& arguably safely), the keto lifestyle eliminates the junk & by limiting the carbs, it also reduces Advanced Glycation End-Product Enzymes. As I understand, no longterm studies have been done on keto but I do understand that it isn't a right fit for everyone. I love how I feel on keto but I cannot sustain it for more than 2-3 months & so now I'm leaning towards the well studied & trusted Mediterranean diet. There are so many factors & variables involved which makes finding the perfect diet very difficult. We all agree that sugar, industrial oils & chemicals/processed is all bad & so maybe it doesn't matter if protein is low or high & likewise, if carbs are low or high - as with fat too... if it is consumed through real food. Fasting can only be a good thing in my opinion because feast & famine makes sense to me. Mindset & a positive outlook, along with sleep, exercise & some socialising/laughter/fun will only help keep disease at bay as we age. Maybe we should all stop overthinking things & just keep it real - as in if it's from the soil, a tree or the sea, it's good enough (organic as much as possible). This is slowly becoming my approach & despite all I've said, I will (still!) most likely choose to have a weekly or fortnightly treat - be it ice cream or an Indian takeaway! Important to have a little something which makes you super happy too! YOLO.

I'm sure the English royalty does not follow a restrictive diet ... just high quality food and a clean environment ... they've outlived all diet philosophies generally speaking

Pescetarian is probably the best low effort diet, but a 100% wfpb diet with the right supplements is probably better, because it avoids all the downsides of animal products. It just maybe hasn't been studied yet.

If I remember correctly, I don't think that diet and bone health study was very good. I can't remember if it was because the vegans hadn't been vegan for very long or if it was because they did not adjust for confounding variables.

@@dana102083 Yes, talking about similar DNA, I saw a study that protein from cows binds to receptor of human cell. But the protein being slightly different, the human cells are being targetted by our immune system .

I think considerations need to be made for high impact and obese as well as we age; all require more protein than average. I think its really diet dependent and how you do it amd your DNA. Its interesting to learn :)

I think protein is a marketing myth. I have been vegan for almost 2 weeks and i feel pretty healthy despite not taking protein or protein supplements. Theres a reason the meat and dairy industry lobby and create funded scientific research to support their products and industty as a whole

It depends on their sport. Many runners eat low protein very high carb diets.

@@EmmaDee I apologize if I startled or offended you May I suggest the following google searches ? 1. castor oil for a beautiful face 2. skin before and after castor oil (click on "images" at the top of page) this one shows women's faces before and after using castor oil on them. 3. has castor oil been around 3000 years 4. will castor oil cure the disease scleroderma 5. Edgar Cayce and castor oil 6. should I buy hexane-free castor oil Put castor oil on one hand at bedtime and then compare in the a.m. It is said to use castor oil on skin only once every 3 days. If you find this info of use, replace "castor oil" first with "glycerin", and then "gelatin" in the google searches. In Europe glycerin is a non-surgical face lift. Models use 2 tablespoons of gelatin a day instead of expensive copper peptides. (it is placed on food before cooking, Walmart has it)

Doctor Valter Longo is getting old too. I am wondering why, now. I am a vegan but I am not getting younger either....and I don't eat junk vegan foods which comes with lots of fats, proteins and grains.

All of us will get old regardless of the diet you eat and the practices we keep. Our knowledge and science is not at a point where we can dramatically slow down aging. Fasting, NAD boosting, exercise, Blue Zone diets, keto cycling, etc . . may reduce disease and improve health span, but when you're old, you're still likely to look old. Tom Cruise and Christie Brinkley do pretty good, but they probably rely on cosmetic procedures. Cosmetic procedures and habits will help with appearance (hair dye, transplants, weaves, plastic surgery, other skin procedures etc . . , sun screen etc. . .) David Sinclair seems to dye his hair every other week. He supposedly claims to have no grey, but likes to change the color with toxic hair dye just because he likes a change every now and then. (I don't believe him. I think he has grey hair. LOL) Chuando Tan looks really good for his age but he admits to dying his hair. If he let it go naturally grey, he would probably look way older. Most older people who look extremely young for their age are likely doing hair dye and / or some other cosmetic procedures.

This is the problem with a lot of longevity doctors. Most look very frail as they age and are not the best looking. I think you have to choose either sexual attractiveness or longevity at some point. Big muscles and curves don’t go with low protein low calorie. Many vegans look wonderful in their youth but it fades.

no one is going to achieve true longevity through eating/supplementation ... sure maybe a few quality yrs ... the only thing will be a real breakthrough, some kind of gene manipulation .. we all know this ... vegan/paleo/keto, etc... accidents and genetic predisposition excluded, with a clean diet we will live into our 80's , give or take a few yrs ... and that's it ... so live clean, enjoy your grandchildren and hope for a gene manipulation breakthrough

I frankly don't see the point of a longevity diet. As an atheist I know that once I'm dead I won't be able to tell if I've lived 5 years or 110 years, so what are these two people talking about here? And if I was a religious person believing in some after life, a longevity diet is pointless, since when I die I go so some kind of heaven. So either way, this is a waste of time. So you're a pescatarian? I'm very surprised. Fish is one of the most polluted food you could eat. Where have you been in the past 10 years? The oceans have become the trash can of our planet. Fish is full of heavy metal, plastic, pesticides and more. There is nothing in fish, if you supplement with omega 3 from algae and iode that you can't get from other food. Besides commercial fishing will collapse before 2050, you seem young, so you should be alive and well, hopefully by then. What will you eat when there's no more fish? Why not put in place a sustainable diet now?

@@bobbobson4030 Presuming Phillipe is young now, life at 80 for him is probably going to be much lower in quality than it is now (less mobility, less brain power, he will be out of his prime and will continue to decline). Perhaps there will be less (and not more) of a concern about extending life at that point.
