Thank you! This was very helpful. Also, if it helps anyone, I got 30 correct based on a guess because A and C are saying the same thing. If the amount of drug bound to the protein increases (A) then the amount of free drug in the plasma would decrease (C). So A & C both must be wrong since you can't have two correct answers. I am glad to actually understand the reasoning behind the correct answer though :)

I have a question about the Ka/ Ko. In order to figure out the binding site for CPFX, would you interpret the value of Ka to be similar to km, on the Michaelis-Menten graph? where a smaller value equals a better affinity for the substrate, therefore the ka for the CPFX-BSA-Warfarin system being smaller would mean, CPFX has higher affinity for site 1. Does this kind of logic work? Please let me know your thoughts. Thanks.

Ok! The good news: I was able to reason through the Ka/Ko and binding site stuff. Bad news: I didn't know that palmitoleic was a fatty acid :P. So big question for the future: If I don't know what one of the answers is/looks like, but I know what the others look like, is there a valid way to reason that the thing that I don't know is actually the correct answer? Maybe I just have an immense fear of being wrong. Thank you for the explanations!!!

Tough question…reason being, I always encourage students to NOT pick the answer they aren’t familiar with. It’s typically a cop-out. However, you could reasonably ascertain that the other three answer choices did not match the characteristics of the structure in the passage. So that, along with “acid” being the root of palmitoleic acid, hinting at it possibly being a fatty acid, should all be clues that it’s the right answer. I think it’s kind of a low blow from the AAMC, but we work with what we got!

@@InformingFutureDoctors OK! Live and learn: if I don't know what something is, but the other options are terrible/not great.....well you only live once. One of my biggest fears as a perfectionist is to get stuff wrong on tests, but I think you mentioned in a video that getting every single question right is not feasible....so next practice passage, if something like this happens again, I will try taking a chance!

Great video, I just cannot square q32 away, I feel like there is a logical jump. How does the degree by which Ibuprofen (Site 2) diminished BSA-CPFX binding affinity relative to the degree by which Warfarin (Site 1) diminishes BSA-CPFX tell you which is the primary binding site of CPFX on BSA? I do not know how you can look at the fact that Warfarin (Site 1) diminishing CPFX binding more than Ibuprofen (Site 2) means that therefore Site 1 is CPFX’s primary binding site. Not sure if I am following the logic correctly.

More precisely: How does the presence of competition (determined by Warfarin diminishing CPFX-binding to BSA more so than Ibuprofen given by their comparative Ka/Ko ratios) lead to the logical conclusion of, therefore Site 1 is the primary site. I would almost say that the fact that CPFX has a tougher time binding to BSA in the presence of Warfarin presents grounds for a stronger case that Site II is the primary site. Obviously this is not correct but I just cannot wrap my head around the rationale for Site 1: "Perhaps it has a tough time at Site 1 (in the presence of Warfarin) because of competition evident by the lower Ka/Ko ratio, therefore Site 1 is the primary site."

@@javierarias-romero2538 Hey! You might have already figured it out, but here is how I thought about it. CPFX-binds to BSA primarily at site I, not site II. Thus, introducing something that ALSO binds to primary site I will be more detrimental to CPFX-BSA binding, compared to if there was a competitor at site II, which CPFX doesn't bind to much anyway.

Like if there was an apple (site I) and an orange(site II) on the table, and I really wanted an apple, I am less likely to get the apple if someone else also wants the apple. But if I want the apple and someone else goes for the orange, it doesn't affect me much.

Ka is the association constant. I’m sure you could find info regarding that online but it’s just like other equilibrium constants. Ko is specific to this experiment but the passage tells you what it means!

Low Km = high affinity. But in this case we’re not talking about Km were talking about Ka, which is the association constant. As with every equilibrium constant it’s gonna be products/reactants and for an ASSOCIATION constant the equation will be A + B = AB. Km is a term relating to Michaelis menten kinetics and it’s the substrate concentration at half Vmax. So high Ka = more association = higher affinity. High Km = need more substrate to reach half Vmax = low affinity. Does that answer your question or am I way off?
