Hello Zach. In the gluconeogenesis section of the video, specifically the part about pyruvate you said that it gets metabolized into AcetylCOA, then enters the Krebs cycle and gets metabolized to OAA, which in turn gets metabolized into PEP through the PEPCK enzyme and eventually into glucose. Based on your other videos I would say the correct pathway is for the pyruvate to be metabolized directly into OAA through the pyruvate carboxylase enzyme and then follow the same steps as stated above. That s because pyruvate(3c) loses a carbon in the transition phase in the form of CO2, and AcetylCOA(2c) brings 2 carbons into the Krebs cycle which are removed once again in the form of CO2 [Citrate(6c)/OAA(4C)], thus not providing the carbons necessary to form PEP and eventually glucose. Other than that it was a great video. Congratulations and keep going!

Great Video. Kudos! But actually, the conversion of the new Keto acids (Intermediates of TCA) is shunted into the gluconeogenesis pathway as Malate and not Oxaloacetate. And this is because, the Mitochondria Membrane has no transporter for OAA. Whereas, the Ketoacids must be transported to the Cytoplasm where most enzymes involved in gluconeogenesis are domiciled. Thank you so much Ninja nerds for your videos. It really help us as medical students. I have a contribution to make. For pyruvate to be used for gluconeogenesis, it is directly converted to oxaloacetate by the action of the enzyme Pyruvate Carboxylase. If I heard you rightly, you said pyruvate can be decarboxylated to Acetyl CoA which can go through the TCA cycle and finally end up as oxaloacetate, which is then channelled through the gluconeogenesis pathway. But we know that as Acetyl CoA goes through the TCA cycle it is lost as 2 molecules of CO2 while the OAA is regenerated. Thank you 😊
