In fluorescence microscopy, we use UV light (or shorter wavelength) to illuminate the sample labeled with a fluorochrome. The fluorescent dyes are the molecules that absorb light at one wavelength and emits lights at a longer wavelength giving a fluorescent effect to the sample.
First, the sample is stained with fluorescent dye. “It is not necessary to stain a sample. Any fluorophore (like GFP) can be visualized with a fluorescent microscope.” The parts of the cells can be stained with different fluorescent dye so as to distinguish between them. FISH (Fluorescent In Situ Hybridisation) employs the use of fluorescently tagged probes which binds to a specific region of the chromosomal DNA.
