Thank you! I am taking Pharmacology and that makes a lot more sense. My professor only talked about Phase 2 Metabolism for like 20 seconds, (possibly because it is an introduction to Pharmacology class), but I still did not understand what Phase II Conjugation was all about. This explains it nicely! :)

What is absorption actually(interms of tablet taken orally),is it the process where drug gets absorbed from gut to hepatic portal system or the process where drug gets absorbed to the arterial circulation? If the process of drug getting absorbed to arterial circulation is the absorption,then Metabolism would occur before absorption,isn't it? So ADME is the wrong order? Could u plz explain it?

@@namonscreen You should listen well to the video. He just said that most drugs are metabolized by the liver or other organs. Absorption during his first video can be done orally, IV or via inhalation. You gotta see the whole picture. Goodluck!

Really easy to understand. Ty for the video. My pharma book explain the basic definition of first pass, none of the specifics, then keep mentioning p450 liver metabolism. I'm so mad they didn't ever explain what p450 does. Now these side effects of drugs, adverse effects, and pharmacokinetics makes a lot more sense. Good thing my final is tomorrow.

Aspirin is hydrolysed by esterase enzymes not CYP enzymes.

When you say "excreted" by the kidneys.. did you mean to say eliminated? Because excreted would imply that it was not used by the body, correct? Or did you purposefully mean to say excreted. My question essentially is this: have these examples been subject to biotransformation or no?

When you say elimination you mean the irreversible loss of the drug from the body by mean of metabolism(which form metabolites from the drug) and excretion (excreted by kidney or in bile), and yes the metabolism of exogenous compounds (drugs) called biotransformation.

Aspirin need to be changed to Salicylic acid. but after Phase I happened, will it not directly go to Phase II? How can salicylic acid come out of the liver before it goes to Phase II?

Oxidation is most common and is RH(drug)O2(molecular oxygen),+ NADPH= ROH+NAD+ + H2O. P450 Cytochrome (heme-protein) Transfers an oxygen atom to the drug RH through the activation of molecular oxygen, introducing one oxygen atom to the drug and one molecule and reduction of the other to H2O. Hydrolysis is not common and happens in drugs like aspirin, which breaks down to salicylic acid and acetic acid. Salicylic acid is the active drug(it was an inactive prodrug aspirin)

Drugs like diazepam, Phenobarbital, chlorpromazine have aromatic rings(like benzene) and these will always be made more hydrophilic with addition of OH group. So oxidation you know from the struture 95% are oxidation reactions.
