To construct a pharmacofor model, is it necessary to know the 3 essential features you describe here? 3:26

The pharmacophore will be different for every hit or lead series, so there will not be a fixed number of features for a pharmacophore. In theory, you could have a pharmacophore with fewer features. I would say that most have more than three. The identity of the pharmacophore features - hydrophobic group, HBA, or HBD - will vary.
