Izuru Kamukura was able to effortlessly develop every possible talent, except hope. He was supposed to be the Ultimate Hope, but hope isn't a talent, and so Izuru chose despair out of sheer boredom. And Makoto is vulnerable to despair just like everyone else, given the right circumstance. When Makoto watched the despair video, he, too, succumbed to its effects, albeit only for a brief moment. Meaning Makoto doesn't have a special talent as the Ultimate Hope; he just chooses to hold onto hope in the face of adversity. Hope is something that anyone can have, and no special talent is needed to conquer despair.

Ultimate Hope is something that Kyoko put on Makoto but Makoto never assimilated it as his own (unlike Nagito who wanted to be called the "Ultimate hope"). Makoto thinks that everyone has hope if they believe in her and that is why he bets on everyone's futures. Makoto is the "incentive" but he doesn't manipulate you or want to control your decisions like Junko. That's why he knows that Junko is a hopeless case (of his own free will it's desperation) but he has faith in the future of Hajime and the other remnants (who are more doubtful because of the wash). Makoto's hope is still the best because it is the purest, many people in real life try to think positive and not get depressed before the obstacles that life gives us, keep going so that the future surprises us, that is the hope that is so exaggerated here. Junko and Makoto are still human, so it is not surprising that the wash takes effect, you forget that Junko also worked "artificial" methods like amnesia and I forget all of her despair in Danganronpa Zero. Both cases only work by being "artificial", naturally hope and despair do not work in them.

I really like how this idea also returns in V3. Team Danganronpa use Danganronpa to try and give hope to their audience, so have always tried to wring out a hopeful ending for them, which can be seen as an artificial hope. That audience can feel the hope towards the end, but for the people involved in the killing game, they don't experience that so much considering everyone they lost. It's when the survivors for Case 6 defy Team Danganronpa and Shuichi, Maki and Himiko survive the wrecking of the Ultimate Academy that a natural hope is born for them and by extension for us, the true audience.

Considering Kyoko's guilt over betraying Makoto in the previous trial despite Makoto trying to save her, I'm shocked she even became conflicted over the decision on whether or not to sacrifice Makoto towards the end of this trial. She should have voted for Makoto without any second thoughts despite the risks Junko said they face If they sided with Makoto.

Note that Makoto actually doesn't says anything beyond his first statement, which is always the same. He doesn't actually tries to convince them further. Heck, Byakuya outright says that he does not need encouragement,he already swore to destroy the Ultimate Despair. SO don't give it toomuch thought,it was just an excuse to make the player shoot somelast truth bullets at the screen.

SPOILERS FOR THE VIDEO AND FOR THE GAME BELOW Come to think of it...Junko had despair ever since she was born because she was bored so easily with things. And what brought her boredom, you ask? Why, her analytical skills, of course! Her true talent, as it's revealed later, is to analyse her surroundings quicker than anyone else, allowing her to build elaborate plans based on her predictions. And these predictions, the sheer predictability of everything around Junko, was what made her get bored with things so easily. It can get boring to win all the time, no matter how elaborate are the games you play. Hence Junko's obsession with despair, the feeling that you have when you realise you've lost. When you can't enjoy your victories anymore, you revel in losses of everyone else. And when that fails, you revel in losing yourself - the one thing you haven't done yet. And this was Junko's master plan - to create the Ultimate Game to lose at! To bet EVERYTHING, to put in a massive effort, only for it to not matter since you lose anyway. For Junko, losing in the Killing Game was like a breath of fresh air. She lost completely and utterly - something that has never ever happened to her before. This was entertaining enough...for the length of the punishment, at least. It's implied that Junko became bored with despair at the very end of her punishment - notice how she changes to a disappointed expression right before getting crushed. In the other words, Junko was so bored and disappointed with EVERYTHING to the point of destroying the whole world to make a game for her to lose at, and even then the result became boring to her soon enough. One'd pity her, if not for the worldwide atrocities she's committed.
