I am 78 and have had treatment resistant depression off and on all my life. I was quite successful in my line of work and only had 2 "bad" episodes until retiring at age 73 and since then, it has been difficult to put it mild. I never responded well to any of the antidepressants and this latest research gives me a ray of hope. Keep up your fine work as unless someone has actually been depressed, they have no idea how much suffering it can cause.

Great video as always. As more research on psilocybin for TRD emerges, I wonder how much overlap we’ll find between patient coverage of ketamine vs psilocybin.

Would you help me find more information about 5-HT2A -2C receptor distribution in the mesolimbic and VMPFC, DLPFC, and Insula-amygdala pathways? And can you tell me the easiest sources, like books with illustrations, explanations, and diagrams (other than Stahl's essential or illustrated books), videos, etc., that help me know more about the different pathways of every neurotransmitter or modulator and their functions in depth?

25mg is too low a dose. You have to use a "heroic dose" of 7 to 10 grams of psilocybin, lay down with eyes covered in complete darkness, no outward stimulus is allowed (eyes covered & no sounds, so no talking to a therapist during the process), a bucket (in case of nausea), & then lay down under a blanket for 6 to 8 hours & go inward where the medicine takes you. It also helps to have a small amount of THC in the beginning & upon awakening from the experience. I got a good six months of complete freedom from my depression, it was immediately upon awakening I realized the black cloud that always swirls around my consciousness was gone. I also quit smoking cigarettes by accident as a side effect. I lost all interest (attraction to, addiction too) in tobacco. I tried to smoke a cigarette the next day & tasted horrible & I put them down without a bit of resistance. But the night & day realization that my debilitating depression has on me was gone. I became a little scared always worried it would attack me again at any moment. But it was a good six months of being depression free & a full year later I was noticing slipping slowly back into the dark fog again.

@@Neuropharmacist you're not supposed to enjoy it. You don't go out dancing on a heroic dose, it's purposely meant to do very intense inner work on the self. Sitting around watching a melting color scheme & giggling, which is exactly what a low dose creates, a low "enjoyable" dose does not do anything to help influence change with regards to psychological issues & neurosis. It's rough taking a heroic dose, it's supposed to be, it's a form of initiation. You drink the tea, cover eyes with an eye mask, lay down in the dark under a blanket, have a bucket next to you in case you become nauseous, & have someone sit with you to make sure you stay safe. A heroic dose of psilocybin makes you sleepy & most people appear to sleep through the experience. But I assure you, the person who is not sleeping, per se, they are on a great adventure & it's different for every experiencer. The mushroom works with whatever you bring to the experience. It can be so transforming that you don't recognize yourself in the next day or two after. I was amazed by how my depression was lifted off of me. I saw the world with new eyes, I was excited about doing the most mundane things & hanging with people that I used to take for granted & actually appreciating them on level that I had never known possible to have. So if you're taking psychedelics for a good time, you're doing it wrong. This is medicine, it's not supposed to be used on a dance floor giggling to yourself, you're missing the whole point of taking the sacrament. Chasing experiences is not a healthy outlook to have on life. You wanna get high & go dancing, smoke a joint & have a shot & go enjoy having fun with your friends. It is disrespectful to waste psilocybin on folly. It possesses above & beyond the ability to shape consciousness towards bettering yourself & teaching you how to devote your existence to something higher than just enjoying yourself with partying & carnal joys. I wouldn't advise you to take a heroic dose, you seem more dedicated to feeling good rather than engaging in the experience of self examination in order to realize what it truly means to be human. It's best you stay asleep & enjoy your low dose of fun, The Great Work is beyond your ability to discern at the moment. Maybe one day in the future when you've exhausted your search for illusions & distractions, you'll come back to the mushroom & you'll be ready for the grand journey to begin. Peace.

​@@humblemonkm61 I think you missed the point. 10 000 mg of psilocybin starts to be close to the LD50 for rodents, which means 50% of animals will die from that overdose. You don't do inner work on 10 000 mg of psilocybin, you go to the ER. You'd have to eat about a kilogram of dry psilocybe mushrooms to get that much psilocybin.

​@@Neuropharmacist I'll give you your 10K mg point because that dose should definitely be worked up to. 7 grams is my preferred dose & I get a full year of no depression from one heroic dose. It's been suggested that I consider doing micro doses as a form of "maintenance" but I don't have a medical facility near me participating in any treatment therapies using that paradigm, when it becomes available i would definitely give it a year & check the results. I'll do anything to keep the Hell of depression from getting a hold of me again. The current mushroom craze revolves around doses at the lower end of the scale - AKA microdoses - which are all about taking a sub-perceptual dose of mushrooms (around 0.1 to 0.9 grams) in a bid to reap the positive benefits. At the other end of the scale, we have the ‘heroic dose’. The term was coined by Terence McKenna, one of the most famous proponents of magic mushrooms, and refers to doses of five grams of the more potent Psilocybe Cubensis species. To put that into perspective, an average macrodose usually sits at around one to 3.5 grams, so a heroic dose is particularly high being anything over 5 grams. “What is usually considered a heroic or excessive dose for most people is anything above three to seven grams, and for some people this means up to 15 grams and for others even more.” So your point is well taken regarding high doses like 10 grams of Psilocybe Cubensis for a psilocybin naive patient. I've done 10 Grams once & a hospital ER was not necessary & have discovered 7 grams is my preferred dose for the benefit I gain. Just review Terrance & Dennis McKenna who dedicated their lives to research & teaching academically about Psilocybin. I'm not pulling information from out of thin air. I've lived what I've shared. Driven to what some might see as extreme measures, but that was only after decades of using Western medicine & slowly realizing how the medical system was more destructive in their medications than one night a year on 7 grams of Psilocybin Cubensis.

I think the word “synthetic” psylocibin versus natural….is significant alters results. Same with synthetic THC……not the same subjective effect at all reported by patients.

@@baran1454 There are some ideas that it could be beneficial, but unfortunately the scientific evidence is still lacking. There are many kinds of strokes and multitude of consequences, so it is really difficult to say for sure. If one was treating post-stroke depression, one could speculate that psychedelics could have beneficial effects based on the previous positive clinical findings related to treating depression. There certainly is a lot of potential for these drugs in the treatment of various disorders, and probably a lot of untapped potential yet to be discovered, but it still doesn't mean these are miracle drugs. I also get a lot of questions asking if giving psychedelics to their relatives with e.g. Alzheimer's disease or Parkinson's disease is going to help. My answer is the same, unlikely, but not impossible.
