I have one question, can we use line and muiltiline discount on single order?

Yes, we can use line discount and multi-line discount on a single order. The line item having line discount trade agreement journal posted will update the net price of that line according to line discount, also if there are combinations present on order which satisfy multi-line discounts, then clicking on "Calculate ==> Multi-line discount" will calculate the multi-line discount for that combo of lines as well.
