In our new video we discuss the main and most important aspects in fixation. Fixation of histological samples is the first and very important step of histological study. It isnecessary to preserve the native structure of the sample. If the tissue is not fixed, the autolysis will start and the structure of tissue will change because of enzymatic decomposition. All the mistakes made on this step will lead to loss of dignostical value of the sample.

The video is very informative and interesting. Please mention which pH of neutral buffered formalin solution is better for fixation and how we will adjust that pH of that solution.

Thanks for the information. its quite educative. However, if tissue (e.g kidney) has been fixed in 10%neutral buffered formalin for more than a month, can it still be useful for histological evaluation? if yes, what are the likely challenges that may occur? Thank you.

You can use any mechanical rotary microtome, for example, Sakura SRM 200 or SLEE CUT4062.

You mentioned 24hr fixation time. What temperature do you recommend?

Biopsy samples do not require 24 hrs fixation , in fact overfixation can be detrimental to tissue analysis

Please, could you clarify the example if 1,5sm = 1,5cm in that way it makes better sense to me.

So I am trying to make some tissue samples. The slides look ok around the edges look pretty good but the inside is just all black, is the sample too thick? Or did I fixate it incorrectly?

Hello! Paraformaldehyde - is a white sediment, the polymer of formalin. For fixation you should use 10% formaline (which is the same as 4% formaldehyde solution).

If I would have stored in 4% paraformaldehyde, can I transfer it to 4% formaldehyde?

@@divyalobi2820 Paraformaldehyde is not a solution, so the sample can not be stored in it. But anyway, use 4% formaldehyde solution for fixation!
