Very nicely done and helpful. I do wish the video showed the rinses being done over a staining tray and properly disposed of instead of being washed down a sink!!

@@HardyDx but if we pour decoloriser more then g postive Bacteria will lose color right ?? So we have to not pour more decoloriser until all the stain comes out ??

I’ve done a few gram stains in the past and the technique is a little tricky just following a procedure. The video helps to show exactly how it should happen.

We're always uses bunsen burner to drying on every step. Will it impact the result?

@vanillaaffogato9985 We recommend methanol because it typically results in a clearer stain. Over-heating of organism on a slide can sometimes cause it to deform or change shape due to heat exposure. Methanol fixed gram positive organisms are also less likely to de-colorize during the staining process.

not a very good technique by the fellow and the method/procedure used its bad. usually you dont attack directly the sample with stain or water or whatever you pour on it. heat its not bad at all, just not direclty for more than 5 sec, like in flames works good enough... air dry could contaminate your sample and/or could have alive bacteria (wich isnt bad at all either but it depends on what you want, generaly dont want alive bacteria so you dont contaminate more tools in your lab).

At @1:25 you've showed that peptidoglycan layer is stained itself. It's not true. It's the cytoplasm that takes up the gram stain.

Hi Sandy! Our manual gram stain kit (Cat. No. GK400A) uses a fast-method, 75% acetone, 25% alcohol ratio. Hope this helps!
