Why do agar slants last longer than just an agar petri dish?

Because of the depth of the agar and prevents it from drying out.

Perfect timing, was planning on making my first slants this weekend. Was curious about using parafilm on the lids. Parafilm is so satisfying to stretch around petri dishes

Until you stretch it too far and it rips I suck with parafilm lol

It makes some mastery using parafilm fast and ensure longevity. It has a few resistance levels, first it's less springy, then suddenly loosens. That's the perfect moment to stick and also ruin it by completely ripping or overstretching enough to get holes there later by itself. Cling film works quite great too (cut roll with a knife into smaller rolls). Just don't mistake for a saran wrap, it's not breathable.

Hello, my name is Sergey. Tell me, do you have experience storing mycelium on slanted agar in a freezer (-20 C)? My experience can be said to be 50/50. Half came to life within a week and the other half did not. I began to suspect that half of the mycelium was lost not due to freezing, but due to drying out. What experience do you have in this matter? The result is the same both on grain and on agar.

please tell me in my country there is no place to buy mycelium, I once bought it from another country, every time it is very inconvenient to buy from another country, what do they do to me please tell me I grow oyster mushrooms, is it possible to somehow store the culture forever and use it? sometimes cloned a mushroom after 2 3 times the strain degraded

Hi Gary, great video! One question. Most of the centrifuge tube lids are HDPE and not PP. Have you had any issues, like melting, when autoclaving them with the tubes?

Hey Gary! Great video as always! I'm going to build a flow hood soon, I'm wondering if yours is made of two stacked filters and it ok to have a seam in the middle? I got a good deal on two filters so I thought I would try it

this one is a single unit with two filters (it’s cheaper to replace two smaller than one large one) but I think stacking would also work

yes if it has the correct nutrients, water content and is sterile/kept from freezing or high temps and uv exposure 👍

how long can mycelium be stored at 40 degrees? I placed petri dishes in my fridge and found that none of them were usable after around 1 year. When I took them out and tried to continue working with them on agar plates, the majority did not grow on agar. Some mycelium did spread a little, but it barely colonized the agar plates. Any advice on what went wrong or why I couldnt revive the mycelium?

I store it in slants in a closet next to my utilities so it’s like 50F in the summer and 40F in winter. I have kept in the fridge for a while but sometimes I worry it could freeze

Hey Gary, I just made some transfers from slants to plates. Slants were stored for around 4-5 months in conical tubes in refrigerator, is it normal for the mycelium from the slants to be a little slow to get going when put back to plates to grow again?

yes I usually take them out at room temp for a day or so and then transfer - this can help a little

@@mintislad3900 +1 for a video on how often you need to return to spore, and the lifecycle, etc. Can you just clone or g2g forever? I've heard folks say theyve used the same culture for nearly a decade without returning to spore, others saying senescence sets in if you dont return to spore (or even if you do). I have and haven’t noticed a difference - I think changing up the food source is more important than the concentration but this is just a hunch.

Why can't you effectively store them in room temperature? Since growth is stalled, how exactly does temperature drop help anything?

Great video!! You make mother cultures from spores or clones of mushrooms? How do you manage cultures for production? Can you make a video about your process?

Thanks a lot of our cultures are from spores but some are from clones too - I usually alternate between pda and mea and start from slants and go to LC and test on agar, use the plates that are clean and repeat.

Too many important points missed and not mentioned here. Philly G.T. explains much better than most, including this channel... He never leaves anything out we might be questioning. P.G.T. Never sticks his face into the camera and instead, does what he's talking about with live examples...
