why did my instructor not flame the loop after each streak phase :( ? i've watched several streak plate videos, they always re-flame the loop after each streak. Should I follow the videos or my instructor?

+Mon Holly One must always re-flame the loop to avoid contamination with other isolation. Your instructor may not be doing different isolation with the same loop ,thats why maybe he/she fasten the process.

+Mon Holly There are two techniques here. Flaming the loop after each streak destroys the microorganisms present on the loop, requiring you to "pick up" microorganisms from the previous streak before going into the next one. Alternatively, you may opt not to flame the loop after every streak. In this case, microorganisms are still present on the loop but in fewer quantities since a large number have been deposited while streaking, thus you may streak the next quadrant, but you must be sure in this case not to cross over to the previous quadrant at all.

I agree, metal loops are much less wasteful. I am not familiar with the 16 technique, but there are many different ways to do it. As long as you get isolated colonies, that is all that matters.
