Hi Ali. Thanks for the question. In my lab, we make our MEM up from scratch, using powdered MEM as a starting point. We are using a CO2 incubator, so we would be using the equivalent of Earl's salts. We always use 10 % serum. FBS is very expensive and not necessary for our cells, so we use BCS. We also add in HEPES & NaHCO3 as buffering agents. Let me know if you want the protocol.
