Sam crying was moving. It was Dean's reluctance to cry until Sam left the room that gave the scene even more power. The big brother always has to hold it together -- even if it kills him to see his sibling in pain.

In my opinion I think Dean cried for the fact that she asked Sam to kill her because she knows there is no way to save her. So Sam made that sacrifice. At that moment it hit Dean that maybe there is no possible way to save Sam and may have to kill him as well

And so many people wonder why Sam messed up so much with the best of intentions. This dude went through way too much. It often goes unnoticed or gets underrated by both fans and the current writers. I hope to see Sam happy by the end of the series. Yes, of course, both brothers should be happy by the end of the series, but Sam deserves it the most. (Just my opinion - I know many people disagree.)
