The short entered the public domain in 1961 as Disney did not renew the copyright as required at the time, making it one of the few Disney cartoons to have its copyright lapsed. However, aspects of the short such as Pluto remain under copyright until 2026. The Mad Doctor character himself was never trademarked and is in the public domain. So, you can use him without permission from Disney.
