Lyrics There was a time when to amplify DNA, You had to grow tons and tons of tiny cells. Then along came a guy named Dr. Kary Mullis, Said you can amplify in vitro just as well. Just mix your template with a buffer and some primers, Nucleotides and polymerases, too. Denaturing, annealing, and extending. Well it’s amazing what heating and cooling and heating will do. PCR, when you need to detect mutations. PCR, when you need to recombine. PCR, when you need to find out who the daddy is. PCR, when you need to solve a crime. (repeat chorus)

For those of you who don't know the story behind the discovery of PCR: Dr. Kary Mullis was out for a pleasant drive in California tripping hard on some LSD. He was thinking about some biochemical problem he was working on. He was visualizing reactions in his head and eventually he "saw" the idea for PCR. Off to the lab he went. God bless LSD.

@MadDragonify You can do PCR with small samples, but there's a limit to how small the sample can be. (It also depends on the kind of equipment you use.) And by degraded, I don't necessarily mean mutated. I mean that over time DNA kind of...falls apart, for lack of a better term. (My professor and I had some problems with that during a research project we worked on last semester...it was very frustrating.)
