Madara awakened his rinnegan using hashirama cells, then put his rinnegan in nagato when he was a baby, then later obito took those eyes after nagato's death, but since rinnegan drains too much chakra he could only handle one eye, unlike nagato who was an uzumaki, a clan known for insane chakra reserves. Sasuke awakened his when he received chakra from hagoromo but he only got one (this part is my theory it's not a fact) because he cannot handle 2 rinnegan just like obito, sasuke may be extremely efficient in chakra usage but he does not possess nearly the same capacity as nagato or naruto. Now the tomoe in his rinnegan are not because it's an upgraded sharingan, no those tomoe are literally a representation of how much chakra he has left, when he runs out of chakra the tomoe dissapear. A rinne sharingan is red not purple like sasuke, which is what kaguya had.

Obito can't handle 2 Rinnegan even with Hashirama cells he can't handle it. AND Hagoromo did give Rinnegan to sasuke, if it was hashirama cells it would have been exactly like Madara's plain Rinnegan. Sasuke's Rinnegan is combo of sasuke + hagoromo. Hagoromo already includes (Indra + Ashura + Toad Sage mode), effectively Sasuke has himself at EMS + Hagoromo + some snake sage chakra, obviously his Rinnegan is more evolved than Madara's plain Rinnegan.
