I think, for Eizen, since killing is the role of the men and the indoor, meant-to-be more comfortable roles were for women, seeing Sagiri choosing to be an executioner is like seeing someone you care about persist in something that's harmful for them, like an abusive partner. She's taking all this pain upon herself when she doesn't need to and having an even harder time than a man would have, while the men are simply put in that role

I think that the job is traditionally for males, so she is an exception, sombody going against the norms. The red hair dude could just be worried, (we even see him stepping in front of her to protect her from the attackers). Regardless of her being a woman or not, the dude is right, the job has a negative effect on her mental health.

My interpretation: Gabimaru is perfectly capable of making super clean kills, he is intentionally making the kills super brutal and messy here to help Sagiri get over her hangups. In Ep1, Sagiri helped Gabimaru realize something very important (that he wants to live + he loves his wife), so in Ep2, he is kinda returning the favor. Just my head canon.

I've read the whole manga and just to be sure rewatched trailer like 8 times by now, all the things happening in the opening literally doesn't make any sense without having the knowledge from the manga, even if you will try to overanalyze everything.

That feeling of misogyny is part of the time and culture, yes. The expectations of a woman didn't include walking the path of the samurai and executing people. I know Sagiri specifically chose to be an executioner to cut off the "middle man" since her whole family business is death anyway but, man, it is quite powerful, narratively, that she'd rather stay in this line work that she deeply struggles with, ethically, than.... getting married off to who knows what. from what we know of arranged marriages it could be a death sentence, after all.

Theres only like one thing in the opening that you might consider a spoiler but it really isnt. all it is is a character with some blood on him. There is blood everywhere in this show as youve probably discovered from this episode and the character in the few frames he is shown literally moves so its not really a spoiler
