Congrats on your successful doctoral defense! 👏🏼 I recently came across João Passos (Mayo Clinic) talk at ARDD 2023 talk and thought that should be bigger news. To actually show that the SASP senescent cells occurs because of leakage of mtDNA and it was possible to remove aging effects by blocking the leakage (dramatically reducing SASP) without killing the senescent cells was quite interesting.

Regarding taurine, you might be interested that inhibition of the taurine transporter (TauT/SLC6A6) mediates muscle atrophy (Warskulat U, 2004; Baliou S, 2020). One cause of TauT inhibition is nitration of its tyrosine residues by peroxynitrate generated in conditions of nitrative stress (Webster, 2008; Khullar, 2004; Roos, 2004). Also, regarding possible selective advantage of aging, there is the theory that where food is limited for a species, individuals of reproductive age compete for food with those past reproductive age, thus hastening death after reproductive age increases biotic potential of a species. Congratulations Dr. S! On astaxanthin, disappointing the ITP used doses orders of magnitude higher than in diet or supplements. Researchers should provide human equivalent doses in their papers to make this clear, and also test realistic doses along side higher doses. Similar for Fisetin in that it appears ITP dose (600 ppm) may have been low compared to what James Kirkland used (20 mg/kg of body weight) or method of delivery (not bolus) not ideal. Again, with fisetin, human equivalent doses should have been given. Simple plasma dilution (Conboy lab 2020 and 2022 papers), a practical intervention, is not getting enough traction as it appears to have many of the benefits as heterochronic parabiosis, not practical as an intervention.

I'm always suspicious of Sinclair's studies in which they claim to "induce aging," and then "reverse" it. Is the repair of artificial damage that they themselves cause really the same as "age reversal?" I highly doubt it. But that's how Sinclair always pitches it. The dude strikes me as kind of a charlatan.

Thanks for putting this together Ms... ahem, I mean Dr Sheekey. I enjoyed the occasional goofy touches (mean that in the most positive sense.. I'm a Pippy Longstocking fan forever lol). If you like constructive critisism, it would be that I'd have loved a conclusion for lay people per section, in some quantitative way that communicates amount of benefit. This satisfies both the deeply technical types and lay people alike, plus .. reaching more audiences is a good way to improve the channel.

What supplements do you take? Are there any special foods that you eat to get naturally formed molecules in lieu of those found in supplements that are extracted or synthesized? Right now I'm taking around a hundred different supplements a day. The one that scares me the most that I haven't taken yet is fisetin. According to Rimon of wellness messiah the recommended dosage is 20 mg per KG of body weight one time once every one to three months. Low levels continuously have now effect which is why the experimental results he cited showed no effect. I never heard of anyone dying of eating too many strawberries. Maybe a stomach ache but not serious illness or death. 😊Would you try it yourself? Have you tried it?

one more you have to know and i found is that senesencce occur because of stem cell deficiency and signals of it propagate to other cells to come to sene and do not apoptosis because there is no there to differentiate raldh1 deficiency as it is the stem cell marker that they express to turn retinal to retinoic to differentiate , beause of this if we use retinoic acid forcefully it will have side effects in long run
