In 30:43 how the answer will be cation exchange resin as our protein has positive charge it will elute out in anion exchange resin .....so answer must be (b) !!!!!
@aaksheebeeАй бұрын
protein is +vely charged so we will us cation exchanger where matrix is -vely charged. protein will bind to matrix and will be eluted last.

@aaksheebee yes,bt wt we want....we want our protein to elute first naa ....
@DerekMohantyАй бұрын
No we want to separate our desired protein from the undesirable one. Once the protein gets attached to the resin or bead and elution is done we will further use salts or change the ph of buffer to wash out our protein.

How can protein having ph 8 greater than pI having positive charge it should be negative
@DerekMohantyАй бұрын
The surrounding H+ ions concentration is greater than the protein hence the protein will get protonated. Thus the positive charge
@BHADRAAMITy2qАй бұрын
No it will be negatively charged... If the pH is less than pI then the [H+] is high in the solution. The protein is fully protonated hence positively charged. When pH is greater than pI then the [H+] in the solution is very low and to maintain equilibrium protein starts to loose all the protons and become negatively charged

😂How am I wrong then!! Have you looked at the question twice?😂 Ph is lower than PI hence protonation hence positive charge on protein. Simple
