So basically: -Laughing your ass off at the extremely witty dialogue that Tina has when suddenly... -4:23 comes....you stop laughing nearly instantly and fall silent... -4:37 happens and you begin to tear up after realizing what its from. -4:37 through 5:27, you're trying not to cry or even make a sound and failing miserably at it. -5:28 returns to its normal comedic tone. You are still in shock. You pause the video and decide to walk away from the computer for a bit and get some fresh air.

The Mission's called Doctor's Orders, it's given by Tanis around the same time you get the story quest to go to the preserve, it has you collect 4 audio logs, I just finished that quest yesterday. Chills man, chills.
