Can i know why is the Plant kept for 3 days in dark(to consume all the starch present in the leaf) & again kept for 6hrs in sunlight, if it is to gain starch then why was it kept in the dark

Hi Aniruddha. Thanks for showing interest. The destarching step in this experiment is done so that the previously present starch should not interfere with the other constant parameters or during the observations. And the experiment is then done under controlled conditions of sunlight, CO2 and water..

Maam what should be the colour of the alcohol solution when chlorophyll is dissolved into it ? Should it be green

since the pigment is very less, so alcohol color will just change slightly to pale side.

This is an appropriate time duration in which the chlorophyll containing parts of the leaf with trap sunlight to synthesise Starch and then we can do the Iodine test to test which parts of the leaf( green or yellow) have Starch

Do you mean that starch is only found in chlorophyll region of the leaf and not anywhere else?

Yes, only the region of leaf containing chlorophyll pigment can trap sunlight to produce starch, in short can do photosynthesis. That's what is proved here in the experiment. Should you have any other questions or arguments please don't hesitate to ask.

@@dime3868 if you are asking about leaf only, then only chlorophyll regions produce and store starch (non green parts of the leaf can't store starch) but there are some storage tissues(non green) in plants like tubers, (potato , sweet potato) and roots of the plants, where a plastid called amyloplast is present, which also produces and stores starch. If anyone sees this comment please answer my question 1. Why does the plant have to be de-starched and then exposed to light again? 2. Why is a variegated leaf used?

I will answer in reverse order. 2. We need to compare the plants response to sunlight at green area containing chlorophyll and yellow area without chlorophyll. Therefore variegated leaf 1. Any experiment is always done in controlled conditions. So we first make the plant utilise all its previous food/ starch by keeping it in dark room. And then we start the test fresh. So now we could fix the duration of sunlight and the time for photosynthesis and hence the time to synthesise starch for every part of the plant equally. In scientific terms we defined the “constants” of this experiment as duration of sunlight, location, water, CO2 content , soil and the “variable” is just one presence or absence of chlorophyll. I hope this will help!

Bhargav jyoti Saikia in board exams u might be asked some conceptual questions based on this topic, like why the beaker containing alcohol is not directly kept on flame, or why do we boil the leaf, or what is the role of iodine and what colour appears after putting the iodine solution on chlorophyll contains section of leaf. Or, it could be a part of objective question, and u’ll have to pick the right sequence of the test. because boiling in alcohol breaks the chlorophyll pigment, decolorising the leaf, so we can then see the color change easily with iodine solution.

Thanks mam but I have a question regarding this the ques. is that if we boil the leaf the starch present in the leaf get removed fully in the alcohol and the leaf get destarched , but when we put some drops of iodine on it, its colour become blue-black at the place where the starch is present , but we have been removed all the starch , now, how the leaf become blue-black ?

Dipak jesthi starch is synthesised by green parts of leaf by the process of photosynthesis, and Iodine is used to test the presence of Starch in the leaf , as Iodine reacts with starch to form a complex which is blue black in colour. I suggest to watch the video again, pause it on each step and draw a flow chart on notebook step by step and this way you will understand the process better

Bhavya Pradhan after removing the previously present starch completely, the plant is again brought to sunlight , and then after 6 hours of sunlight the test begins on the leaf.

Monisha G. S. This is a very simple test, only Iodine is sufficient. Although sometimes we don’t see the result as clearly as it is known theoretically.

Adyasha Das because we want plant to synthesise starch in the controlled conditions that we are providing, and then we test which parts of leaf ( green or yellow) have synthesised starch using Iodine test.

Because green colour of chlorophyll could interfere with blue black colouring test of Iodine solution on the leaf. With dark green colour present it’s hard to observe if the leaf has changed colour on adding Iodine.

M Shashank . So the section where starch is present, iodine solution turns to blue -black ( forming a complex with starch), while the section where there is no starch, the iodine drop will retain its orangish colour...and no colour change is seen.

Gaurav Pandey . The leaf is boiled in alcohol to remove the green pigment , chlorophyll. So that the color change is distinctly visible at the end of the test.

Insane Gamer only the green patches of the leaf had the chlorophyll which was also later removed after boiling in alcohol, to test that which patch of the leaf was successful in synthesising the starch.

Can i know why is the Plant kept for 3 days in dark(to consume all the starch present in the leaf) & again kept for 6hrs in sunlight, if it is to gain starch then why was it kept in the dark

Naveen Sundar we are doing this experiment to test if chlorophyll is needed for photosynthesis or not. we need to remove all the previously present starch, So that we could validate the synthesis of starch in green and yellow part of the leaf during the experimental 6 hours of sunlight. In layman’s terms, to get the precision and authentic proof of starch synthesis in only the green portions of the leaf.
