This is a very interesting topic and one that is very dear to my heart. But for the 99% of people who are not going to do this at home, I have had success with an alternative that doesn't require aseptic condition: for any plant that can be grown emersed, you can take small cuttings with at least one node and grow them in small terrestrial plant pots of potting soil inside a plastic bag (which is at 100% humidity) under medium lighting conditions. In the event that you don't have much material to work with or the plants are dying, you can increase your success rate with a dip in rooting hormone powder. In plant organogenesis, auxins produce root tips, which then produce cytokinins for shooting, so exogenous cytokinins may not be necessary. The major advantage of tissue culture is that very tiny explants can be used because the plant does not need to have an immune system in aseptic conditions.

Can you put the explant in clonex and put it in a bag with soil? I really don't have a full idea of this I just take regular clones and use clonex the old school way.

@@manicmurph Tissue culture is bascially taking a section of a plants node and putting it in PERFECT conditions to grow. Nodes have stem cells which can specialize into any type of cell (stem, leaf, root etc., basically they haven't made their mind up yet). Taking a cutting the traditional way requires at least 3 nodes to have an acceptable success rate which is usually around >60% successful (>80% if using 4 or more nodes). Tissue culture can technically be done with just A SINGLE CELL from a node with a near 100% success rate. The only issue is the risk of contamination, but if you follow sterile procedure, you can literally turn one plant into thousands, which can then be turned into millions... If you want to learn more about this, I would recommend the youtuber "plants in jars". You can even do tissue culture with liquid media if you don't want to deal with the jelly media in this video.

Man this is by far the best culture medium video explaination I have seen. You broke it down such an easy wording that anyone can follow. Thank you so much. You have inspired me to take the leap to tissue culturing of plant. I mean I have been propagating plants by stem cuttings, water culture, and misting. This seems like the nature course to take, so thank you.

What a great presentation (minor blips aside). There are so many awful presenters that say nothing and lots of it. I like that you didn't tell us things like, what you ate for dinner, what your dog ate for dinner, what color your underwear is, nothing that wasn't relevant. So many presenters could learn from that alone.

why does it have to be that complicated? Can't I just make a system that fertilized water flows bellow the plant to get the nutritiens directly from water? Like a pump running water bellow the plant which is either in a spunge or on a wire mesh? Mainly I am planning to grow carpet plants. So, it is a challenge to grow like this but is it possible?

Are you using plastic cups? How are they not melting when you pressure cook them. What kind cups are you using? I don’t see the link to purchase ppm or the other ones you mentioned.

I use cups advertized as slime cups on amazon, they are pp5 or something. Its kind of like hardness grading for plastic

Polypropylene aka #5 plastic doesn't melt at high temperatures. Commonly used for food, medical, and scientific purposes. Most #5 containers should be sturdy enough to sterilize multiple times.

Can you tell me how to make the plastic box for growing the tissue culture? Do you keep a fan blowing in it at all times? Do you have to sterilize the fan?

You can just grow them in a sterilized plastic tote. Technically you can grow them on a shelf if the containers are air tight. (Which they should be)

Mirak Subba, is there a reason you can't propagate a bunch of cuttings. Or do you want a ton of plants? I see no reason why this wouldn't work for philodendrons, although you would probably get better results with different levels of plant hormones. One thing you could try that would be simpler is to spray your plants with just the hormones with a high BAP ratio to produce more shoots then propagate cuttings in a high IBA ratio solution to encourage root growth.

The bleach available in stores is diluted so customers don't commit accidental retroactive self abortion the moment they open the container. Some brands have been known to carry more intense bleach so if you try a new brand 20% could potentially damage the plant. 5-10% should also be fine.

You just adjust pH once at room temperature. If you rise the temperature of the solution the pH will drop but once it cools to room temperature pH will return to the same value, unless there are some very volatile elements on it. For instance pure water has a pH of 7 at room temperature, bring it to 100C and pH will be 6,14. Let it cool and pH will return to 7. Nice video. Have you tried to grow on liquid solution? By the way, I wonder how your containers don't explode inside the PC with pressure. In my case the lid is blown so I had to figure out another method.

Sorry to be a nitpicker but what is 20% bleach??? Are you saying 1 part Clorox brand bleach for 4 parts water? That would be roughly 1.5% sodium hypochlorite which would melt any plant in about 2 minutes... how are you able to do this for 10 minutes? The plants should be totally white.

Hello 10% bleach is more widely used so yes 1 ml of bleach per 9 ml water. Some plants are more fragile than others when it comes to bleach

Merry-stem culture is very easy, you just need that microscope. You don't need to be a scientist to dissect out the merry-stem, it's fiddly but DIY tools are just fine to bump and scrap it free particularly as most aquatic plants are so brittle. The plus side to using meristems instead of other plant tissue is that your plants will be guaranteed virus free, plus of course you need very little material. It doesn't work with waterlilies or we could already have a truly "blue hardy" one by now, the first was lost during a flood in Thailand, sterile and refusing to be propagated by any other means it probably ended up floating around in the Andaman sea.

Why make a still air box if it's not sealed? Can't contaminants just float in through the holes? And you really shouldn't rely on sterilizing your hands as they're porous. You should use gloves if you're not gonna keep the box sealed 1000% of the time.

You dissolve it in a seperate container at a 1ml/mg ratio , so you have a stock to pull from.
