HIND I is first exonuclease restriction enzyme HIND II is first endonuclease restriction enzyme thats why first endonuclease is given roman number II because exonuclease and HIND II is first endonuclease

Roman number does not indicate the order of discovery. It indicates the order of isolation of the restriction enzyme from the particular strain of a bacteria. Since Hind-II was discovered earlier than Hind-I, that's why the first discovered restriction enzyme is Hind-II. But Hind-I was first isolated from that strain of bacteria from which Hind-II was isolated later. That's why there's II in the first discovered endonuclease enzyme.
