And when the reading is 24-25 volts on alternator output but 12.1 volts across the battery, are we looking at a short in alternator or something wrong with the charge controller? 80A alternator for Lexus ES300 yr. 2000

I didn't understood. There was a big difference between the two test at the end. What does that mean?

but according to ericthecarguy, the reading should be 0.01-0.02..if the reading is 0.10-0.20, then there's a problem.

Actually if you go back and watch the ErictheCarGuy video again, he says that the voltage drop on the negative side should be between 0.1 and 0.2. No numbers on the positive side.

and different cars have different specs alldata tells you the range it should be, generally under .50
