Why do we have to have to separate proteins by weight in the first place if we are just gonna use antibodies to find the protein of interest?

Imagine the proteins aren’t separated and they are at a single band. Try to probe for them, you’ll just get a large blob of any-protein. You won’t know what protein is what. Separating proteins of interest by weight allows for the probing of specific proteins of interest, so long as the protein on one gel is not the same size as another protein of interest you would like to probe for. You would run and probe for that second protein of interest on another gel. Hope this helped, lmk if anything isn’t clear or if I said anything incorrect

@@ala.9810 yes you are correct. If the proteins are not separated at first place then the enzyme or the antibody will not be able to bind properly, so you cannot detect the protein of interest. Other proteins if not separated might inhibit the attachment of antibody. Thank you .

Thank you for the video! you explained in 6:31 what my lecturers couldn't explain in an hour. the one question I still have is: what is the point of SDS in western blot? surely you can skip SDS because you're using Antibodies, why do you still need to separate the proteins based off molecular weight? thank you

I think you have to separate the proteins because if you didn’t there would be no way to identify your protein of interest, it would just be one band. Separating them by size gives you something to compare your protein ladder to, which has a standardized banding pattern with known sizes for each band

I believe using SDS-page separates a sample into proteins based on their molecular weight (kDa). This allows for protein isolation, especially after the gel is moved to a membrane that can withstand the washing during antibody binding. If we used just the antibody technique, they likely wouldn’t bind to anything since antibodies are highly specific to a certain sequence of DNA. A sample may contain material clumped up into various secondary, tertiary, quarternary structures with the sequence of interest covered by all the other DNA.

vertical gel electrophoresis, the gel is vertically oriented and the buffer system is discontinuous which is important for the stacking gel and running gel.this is not possible with a horizontal setup
