Why do we have to have to separate proteins by weight in the first place if we are just gonna use antibodies to find the protein of interest?
@ala.9810
@ala.9810 2 жыл бұрын
Imagine the proteins aren’t separated and they are at a single band. Try to probe for them, you’ll just get a large blob of any-protein. You won’t know what protein is what. Separating proteins of interest by weight allows for the probing of specific proteins of interest, so long as the protein on one gel is not the same size as another protein of interest you would like to probe for. You would run and probe for that second protein of interest on another gel. Hope this helped, lmk if anything isn’t clear or if I said anything incorrect
@manabsenapati3996
@manabsenapati3996 2 жыл бұрын
@@ala.9810 yes you are correct. If the proteins are not separated at first place then the enzyme or the antibody will not be able to bind properly, so you cannot detect the protein of interest. Other proteins if not separated might inhibit the attachment of antibody. Thank you .

Thank you for the video! you explained in 6:31 what my lecturers couldn't explain in an hour. the one question I still have is: what is the point of SDS in western blot? surely you can skip SDS because you're using Antibodies, why do you still need to separate the proteins based off molecular weight? thank you

Good question....I will answer you tomorrow morning ( it's midnight in India)
@Rectanglesful
@Rectanglesful 2 жыл бұрын
I think you have to separate the proteins because if you didn’t there would be no way to identify your protein of interest, it would just be one band. Separating them by size gives you something to compare your protein ladder to, which has a standardized banding pattern with known sizes for each band

@@animatedbiologywitharpan Are you working on another video for them so clever. I'm not on this level yet. But always interested to learn more. Do you know has any one in the entire world yet isolated 2019n-CoV and come up with any real images and not simply animation? I have been searching from the time it was announced, back in early 2020. However, Dr Peter Daszak reiterated that, "until an infectious disease crisis is very real present, and at an emergency threshold, it is often largely ignored. To sustain the funding base beyond the crisis, he said, we need to increase public understanding of the need for MCMs such as a pan-influenza or pan-corona virus vaccine. A key driver is the media, and the economics follow the hype. We need to use that hype to our advantage to get to the real issues. Investors will respond if they see profit at the end of process, Daszak stated.” Dr. David Martin, shared that quote, and also this one; "The SPIKE PROTEIN in the injection is a bioweapon defined under U.S. Law 18 U.S. Code § 175 - Prohibitions with respect to biological weapons (a) IN GENERAL. - Whoever knowingly develops, produces, stockpiles, transfers, acquires, retains, or possesses any biological agent, toxin, or delivery system for use as a weapon, or knowingly assists a foreign state or any organization to do so, or attempts, threatens, or conspires to do the same, shall be fined under this title or imprisoned for life or any term of years, or both. There is extraterritorial Federal jurisdiction over an offense under this section committed by or against a national of the United States."

I don't really understand the use of this technic. Why use SDS-Page ? We could just only use the antibodies technic ?
@Casualbystander
@Casualbystander 3 жыл бұрын
I believe using SDS-page separates a sample into proteins based on their molecular weight (kDa). This allows for protein isolation, especially after the gel is moved to a membrane that can withstand the washing during antibody binding. If we used just the antibody technique, they likely wouldn’t bind to anything since antibodies are highly specific to a certain sequence of DNA. A sample may contain material clumped up into various secondary, tertiary, quarternary structures with the sequence of interest covered by all the other DNA.
