Unless if you're implying sibling marriage (guaranteed to be 1/2 Uzumaki since both parents are half-Uzumaki) or the mother is at least half-Uzumaki (from a different branch) then there is no way Tsunade is half-Uzumaki. Naruto however is half-Uzumaki, since Kushina is pure-blood while Minato is not an Uzumaki.

The Uzumaki clan and the Senju clan are related directly by blood that's why both clans have better bodies and more chakra than the average shinobi because they directly inherited the Sage body from the Sage of six paths through Asura even if it's only small amount of it but it's enough to increase the amount of their Chakra to unnatural levels compared to other Ninjas, and also this is the reason Asura's reincarnated in this two families unlike Indra who only appeared in the Uchiha clan. So actually the first Hokage with an Uzumaki bloodline is Hashirama Senju himself and later Tobirama Senju.

you could just read the manga. but essentially, they come from the former whirlpool village that went extinct. migrated into the konoha village, Mito married Hashirama, had a kid, kid has Tsunade, most of them migrated into other clans across the world and that's that. the jinchuuriki was just around and hashirama captured kurama by just hunting. however Uzumaki had the power to host it and chose mito as the first host. they excelled at sealing jutsus.
