So is the transgene delivered in the adenovirus or the AAV? is the only genetic material in the AAV the rep and cap proteins?

Hi Gabriel, the video describes AAV as it was discovered and its interactions with Adenovirus. When we are looking at using packaged AAV virus carrying a transgene for transduction, the genetic material carried by AAV consists of the payload (typically a promoter and a transgene) flanked by the ITR sequences. The Rep and Cap proteins are expressed from another plasmid during packaging, but this plasmid does not get packaged into the AAV itself because it lacks the ITR regions which are only on the plasmid carrying the transgene. This provides an additional level of biosafety, requiring the packaging plasmids to be present in order for the virus to replicate.
