No, only the Companions and Keepers need to do that shit. Through i think Keepers do have to patrol from time to time Normal Custodians move around to perform their duties. There are reasons that those positions require the best even among the superhuman warriors of Custodians. Even for them, sitting still for years, decades or even a century while ready to spring to action at anytime is not easy. Add on top of that are the psychic inferno around the Throne or hellish conditions in the Dark cells. The Captain General and Keeper Captain need to mindful of rotation

You left out the fact that the custodians that boarded the vengeful spirit with the emperor were actually tainted and turned by Horus, and they not only attacked the emperor but he was forced to kill almost all of them that had accompanied him! So yes not only did he have to fight his son who was all warped Juiced up, but he had to fight his own bodyguards so ain't that just a bitch? The custodes only "weakness" in combat is psykers and demons. They're extremely resistant to psychic power with marine librarians struggling to even slow them down, describing it as interacting with a wall of indomitable will, and are often deployed with sisters of silence to level the field. Tanau Aleya in watchers of the throne says "if it bleeds a custodian can kill it" and sisters job is to anchor demons in real space so that they can be slain

They did venture out previously with the dread host specifically dedicated to destroying threats before they grow and the Aquilan shield protecting key individuals who have some role to play. They weren't all 10k on Terra all the time.

I've heard someone say the custodes can replenish their ranks, is this true? Cause that makes these guys kinda busted. They are generally considered to be the best of the best in the 40k universe and outside of a couple times we've only heard of them as unstoppable forces. Send one at Abbadon and genuinely believe they'd be a match for him. If a single chaplain of the blood angels can land one clean hit against Abby, then I assume a custode(If their abiltiies are true) would genuinely challenge him.

Custodes recruitment begins in infancy with noble families offering their children through the avenue of sacrifice on Terra. The process of their creation is not known beyond the walls of the palace but is said to border on the occult as they have the Emperor's genes within them and the transformation takes place in the cells. With no surgeries for additional organs akin to the astartes. The process is extremely precise but very costly. Before the opening of the great rift they numbered at 9842 I believe, just under full strength according to "Watchers of the Throne". TLDR: they can, it's just extremely fucking costly and takes a long ass time

It's not like marines where they can just have a bunch of aspirants and extra on top to account for failures and deaths before they finish the trials and recycle the organs later.
