As per Gene Cloning & DNA analysis by T. A brown 6E(P111), the presence of the insert DNA in the vector can be checked by testing for insertional inactivation of SUP4, which is carried out by simple color test, white colonies are recombinants & RED are not recombinants. While as per Analysis of Genes & Genomes by Richard J Reece: YAC in chapter Artificial chromosomes RED colonies are recombinants. Now what's correct

Red are recombinants. Insertional inactivation of SUP4 gene prevents suppression of ADE1 and ADE2 gene mutations. So now they will get mutated. Adenine synthesis will get interrupted and a red intermediate accumulates.

@@Navya20046 I guess it's because linear vector resembles Eukaryotic chromosomes which are linear and have telomeres at the ends. Maby if we wanted to use it in mice it would have to resemble mice chromosomes to work properly.

I found intormation thet a circular YAC vector is able to replicate in E. coli due to the presence of bacterial ori and bla gene and is able to propagate in yeast as a linear molecule containing all necessery chromosomal elements: CEN4, ARS1 and two TEL which are functional in yeast after linearization.

2:37 In real life this insertion (ligation) part never works. Instead you spend days to weeks troubleshooting. That's why I moved on to more efficient but slightly more expensive cloning methods like NE HiFi and Gap Repair. Of course, science being science those don't always work either. After 5 years in the lab and a worthless masters degree, I decided to go to medical school. Science wasn't worth it for me. I would've lost my mind if I had become a PhD student.
