Nice explanation. You have mentioned epitope simply as they are present at the surface of pathogen. That's for sure it's a part of antigen. But I think more precisely it should be defined as the part of a antigen that binds to paratope of an antibody. Paratope is the part of Fab region of antibody. More precisely the Complementarity Determining Regions i.e. CDRs. But yeah it's a very nice explanation.

Nice explanation ma'am. I have a question like for negative case you said that before adding substrate we do washing step but if the capture antibody and secondary antibody has bound to antigen but we don't know that as solution is transparent so at that time we also do washing or not like you didn't mentioned washing step in positive case before adding substrate like logically we should do washing. Can you please explain this?

The antibodies used in either direct & indirect sandwich method are genetically modified for particular antigen, Also this method is for antigen detection so only antigens are taken from patient's serum not antibodies.
