Interesting procedure.I prepare many plates in my laboratory for all bacterias blood agar,nutrient agar , Maconcey ,Endo DC agas ,SS agar and I prepare agar tubes KIA or TSI tubes agar

What's the point of using a hood without using a glove. How's that aseptic technique.

You may be right - the clean bench is not necessary for making sterile microbial media, neither gloves (note: gloves are not sterile)! A clean working place and a Bunsen burner flame are sufficient for production of uncontaminated agar plates with microbial media, though I would not recommend it for cell culture media. Care has to be taken, of course, not to touch any surfaces that may directly or indirectly come into contact of the medium.

@@broadcastbiology9237 I mean…technically they will be free of growing bacteria after being autoclaved. However gloves ARE sterile if you buy them sterile from a manufacturer. And there is a reason that under GMP/GLP you use gloves to prepare agar plates. The burner prevents floating germs from reaching the plates which has near zero impact compared to human fingers fiddeling around there.

how do i know that how many plates are required for the pouring of agar?

Depends on your experiment

U need 15 to 20 ml of media to pour one plate on this basis u can calculate
