Japanese and American humor is generally pretty compatible since both focus on absurdism. The main difference is Japanese humor leans more towards being “weird” while Americans lean towards “crazy”. It's referring to the other Conan, the detective Conan, since he "should" be the only Conan. It's a joke, I'm sure they understood it with the context. Also, it's japan where the only true god concept isn't really that significant and people aren't insulting your religion constantly that your god is "wrong" god. If you did that in places like Middle East, or India where religion is a sensitive topic and people have been insulted with the "only true god" concept, I'm sure people wouldn't be so comfortable laughing at that even with the context, and even if it came from Conan. But I'm sure Conan is smarter than that and would take it into consideration before entering the country or engaging with the people.
