Very easy to follow explanation. I think you could have made it a little bit shorter, since only the last 5 minutes are important for understanding the in situ approach, but when you fast forward through the video it's a quick way to relearn the principles. Helped me for my exam, thanks!

Why the mask and blocking agent both are used? Both have the same purpose not to add nucleotide.. Let suppose why we can't only use mask as mask will also not allow the nucleotides to bind with grooves.. We have to remove the Blocker everytime before adding any nucleotide.. then Why we can't skip Blocker? And 2nd thing why blocker is added with every nucleotide which we are adding in the grooves.. Kindly make it clear plz

How those machines function? From what I read process is similar, although they deprotect with chemicals rather than laser, but that is not my area of expertise :) so I may speak gibberish xD
