Correct me if I am wrong, but I was looking through the Edvotek 301 lab and based on the EcoRI digestion sites, wouldn't a single insert in either direction result in the same fragment sizes of 3000 bp for the vector ad 1282 for the insert. Determining orientation of insert would not be possible with gel electrophoresis. In the video, at 1;57 if the blue region represents the insert, there is a digestion site for for EcoRI that is within the sequence of the insert and not at the end (I believe this contradicts the sites found in the Edvotek 301 manual.) Thanks for your help with this matter.

Hi! Great catch. I can see why that was confusing. The complex plasmid map was show as an example of a plasmid map, but the actual restriction digest performed and analyzed was a simpler one that could be used to show directionality of an insert. As it says in the video description, the plasmid used in this exercise was not for commercial sale.

Why is it called DNA Ladder? Is it just because it looks like a ladder? I know nothing about biology or science (and I don't know why I'm watching this video 🤣)
