Extracting caffeine from coffee

We will be extracting some caffeine from coffee today! I used regular coffee grounds, but I think the process is actually faster and easier with instant coffee.
In theory the yield should have been hovering around 300mg if ALL the caffeine was removed. However, I got around 10% of that (30mg). Apparently 10g makes about a cup of coffee, so a yield of 30mg isn't that horrible.
Industrially, the caffeine is made synthetically and not extracted because it isn't really economical. The only time caffeine is extracted is for making decaffeinated coffee. But in that case, the end product is the coffee itself and the caffeine is a by-product. It is just much cheaper to make the caffeine directly.
Ill be doing a future video on how to get caffeine from caffeine pills.
I am lazy and it is getting there. I have been spending most of my time focusing on filming new experiments, so I often forget about the other things.

This morning, I performed a caffeine extraction from coffee beans. I didn’t have a mortar and pestle so I used a coffee grinder-seemed to work. Next, I placed a coffee filter in a funnel and dripped water at about 100C through it. I left the product on a hotplate to reflux for about 10 minutes, then I poured the solution into a ceramic beaker (that has the periodic table printed on the side). Next, I added about 10 grams of sucrose and a cold buffer solution of lactose, casein calcium phosphate, and lipids. It was good… Seriously, I've watched a lot of your videos and you really have the hands-on organic lab skills. I particularly like the fact that you admit mistakes and paths not taken that might have worked better and the fact that you can make or clean up hard to find chemicals. Very well done and enjoyable to watch. Much respect.

One thing that I would suggest to knock off some of the preparation for this extraction procedure is to take the coffee grounds and the sucrose and dissolve it in about 10 mL of water, this helps with dissolving it in the lactose solution quicker.

Two points I gotta make: Don't be afraid to use extra water for the coffee bean extraction- you can always reduce down by boiling (caffeine is pretty hard to burn, you'd have to get it past 235C to even think about it). Also I feel like drying the DCM was largely unnecessary and even harmful (the salt water likely stole a significant portion of your yield). Since you are drying your solvent with heat, why worry about having an anhydrous solution?

Did this at school in the 1960s but used a large catering can of Nescafe instant coffee. Got quite a lot of caffeine yeild out of it which went very well with the moonshine alcohol made from fermented and distilled sugar and yeast. Hell of a party that weekend.

DCM is an organic solvent, think of this as if you were to dissolve a bunch of salt in water, then it's intuitively evident in order to get rid of the solvent you'd boil or evap the water and in the end you'll be left with salt

Nice video! One thing you might want to look into is to simply grind finer. In the coffee community the consensus seem to be that after a certain size only the exterior gets extracted - that could explain the low caffeine yield.

@Eric Johnson that's why I edited the comment after realizing this hiccup. It could be used the same if dilute enough although it would no doubt taste unpleasant. Remembering back alkali/alkaline earth metal carbonates are a large constituent to ash from wood fire. They are usually a bit less soluble than the bicarbonate form and may have higher ph though I never got to test that.

I think part of the problem in the low yield may be that the brewing technique that you used didn't get all of the caffeine out of the coffee. I say this because it has been found that properly brewed pour over coffee (which is effectively what you were doing) has a higher density of caffeine than espresso.

Nice video! We compared caffeine content years ago at varsity on different beverages. Contrary to popular belief, a shot of espresso had the lowest caffeine content of everything we tested (lower than tea, instant coffee and perculated coffee, etc). This was mainly due to the short infusion time when extracting espresso from the machine (20 seconds). The type of roast also influenced the caffeine content - the darker the roast, the stronger it tastes, but the less caffeine it contains due to longer roasting. So if you want to stay awake, leave the espresso and opt for a weak tasting, but "powerful" pale roasted perculated coffee.

It might have helped to extract the coffe powder in mild acid. This makes caffeine more soluble in water AND prevents the roast products from solving in water. Then alkalize to deprotonize the caffeine and extract with CH2Cl2.

To expedite the filtration and minimize coffee loss I would recommend dampening the filter with DI water. Wet it after you begin brewing and it should be damp but not soaked.

Copy paste Put in 10g of coffee into a container followed by 4g of sodium carbonate, then add in 60mL of distilled water (increase amount of ingredients for larger quantities) , and then bring the solution to boil for 15 minutes, stir or shake occasionally to prevent it from foaming over, or turn down the heat to simmer, once the 15 minutes is over you can let it cool but make sure to keep the temp as high as possible, and then strain it in a coffee filter, and then add 100mL of boiling distilled water, then pour the mixture into a separatory funnel and then add 15mL of dichloromethane, and gently shake it around, after that pour out the DCM which has the caffeine in it, and repeat the process with the remaining coffee, and then pour the DCM into a clean separatory funnel along with a saturated salt solution to pull water from the solvent, and then gently shake it again before pouring out the DCM, and add in drying agents like molecular sieves and dry for 20 minutes before placing it into a new beaker, and the boil to remove the DCM, but don't overheat it as it will burn, and then, once dry, recrystallize the remaining substance with a 95% ethanol solution, and then let to rest in the freezer before vaccum filtering and letting it dry in the vaccum for a few minutes.

You should try this experiment again with chloroform as the solvent. I am planning out the experiment and still have to make more chloroform. Caffeine is apparently 2 times more soluble in chloroform than in DCM. The only downside would be the fumes from boiling, but you have a fume hood. I think it'd be interesting since basically everybody uses DCM for caffeine extractions. Well, I tried it with caffeine pills and it worked really well. I got a relatively low yield because I was sloppy and a lot of the caffeine was lost on the walls of the containers after the chloroform evaporated off. I didn't have enough chloroform to do proper washings. Still ended up with about 8 grams of impure caffeine. I'm going to recrystallize it from water tomorrow.

+fpm1979 I should have mentioned that! If the goal is to get caffeine, they just make it. If the goal is decaf coffee, there are several. ways. Two commons ones are super-critical CO2 and using methylene chloride directly on the coffee beans.
