This video is just to orient you to the Fanconi Anemia Pathway and is NOT the most up-to-date in terms of the molecular pathway. Review paper used for this video: The Fanconi Anemia Pathway (D'andrea and Rodriguez 2017): www.cell.com/current-biology/comments/S0960-9822(17)30947-8

Hi! This was very helpful, would you be up to make a more in depth video specifically for the initiation stage of the FA pathway or do you have a very good review? I have been trying to find a good review but there is not a really nice up-to-date one.

Hello Kate, Thanks for taking the time to comment on this. I haven't got a review to recommend but here's a paper from one of the leaders in the area, where you can potentially access useful references: pubmed.ncbi.nlm.nih.gov/22279085/ Have a great day.

wanted to know which biomolecule is being affected in this disease

There are to date, 23 genes (nucleic acids) and their protein products (polypeptides) in the pathway. The genes and their protein products contribute to repairing DNA when it is damaged. The genes also play roles in assisting during DNA replication, transcription and in other cellular processes. The 23 proteins also cooperate to remove interstrand crosslinks that are caused by either external or internal factors. In this way, the FA pathway contributes to maintaining genomic integrity. If both copies of any *one* of the FA genes inherited from mum and dad contain a mutation, then Fanconi Anemia disorder manifests.

Hey, I think you say BRCA2 instead of PALB2 here: 12:14 Thanks a lot, great video!

Hi thanks for taking the time to watch and give input. I mention the full name: Partner and localizer of BRCA2 (PALB2). That's why you heard BRCA2. Are you working on these proteins?

@@adwoabiotech Ok, didn't know that. I came across your video because I had a presentation to make (University) about an article that focused on FA and FA pathway
