Nowadays most labs use automated cell counting systems to determine the number of cells in solution, although the classical counting with the Neubauer counting chamber is still in use and it is rather easy to work with it.

Thank you for the presentation however your explanation at the 2 min and 27 mark about which cells to count and what shouldnt count isnt entirely accurate. I had to listen to it a few times but counting the cells in the blue boundary (bottom and right) is inaccurate because you dont know if it truly falls within that square. However if you are counting cells at the top and left, there are no other squares on the outside of those boundaries so it is safe to include those cells in the estimation. I am open to understanding this more and look forward to your response. Thanks

He counted 5 large squares ( 0.1uL each ) and counted a TOTAL of 50 cells. The average number of cells per large square is 10. but each large square is 0.1uL. We want to know the average # of cell per mL but we have 0.1microLiter. We need 1000uL to have 1ml. To get this multiple 10^4 to 0.1 microliters to get 1000microliters. Now, to solve how many cells are in 1 milliliters. You have to remember that solution had been diluted by a dilution factor of 10, and also we have 10 cells per 0.1uL. So it will 10 cells/uL x 10 dilution factors x 10 ^4 = gives us number of cells per 1000uL as opposed to 10 cells per 0.1microliter.

Wait i saw a different video saying u can't count the cells on the lower and right corner lines but u are saying u should im confused now but that guy was using some trypen blue method which he called it

It isn't important which of the corner lines you don't count. You just have to choose two touching lines and stick to that while you're counting to prevent you from counting cells two times.

Hi! Thank you for the video! I have a question regarding the cell counting, cause in some labs they only count 4 corner squares, but here it says 5. Does it depend on a cell type? For example, should I use your method in counting HEK 293 cells (Human embryonic cells)?. Thank you in advance!

Short answer: The more squares you count, the better... It is totally fine to only count 4 and it also depends a bit on the cell type and the quantity of the cells in suspension. HEK293 cells can be counted with a Neubauer counting chamber! The only important thing in general: Always make sure your cells are not clumped together or aggregated. As long as the cell type is visible under the microscope and as long individual cells can be separated from each other, any cell type is fine to be counted with this device!

Technically, you could also count all cells in all squares. But: You have to consider that in the calculation later (divide by 9 instead of 5 to calculate the mean).

The counting is not right if you count diluted cells. you have 100000 cells per ml and the total number of cells is: 10*100000 cells making a total number of 1 million cells in 10 ml

power 4 = multiply 10 to itself 4 times , i.e. 10 x 10 x 10 x 10 (10,000) power 3 = multiply 10 to itself 3 times , i.e. 10 x 10 x 10 (1000) power 2 = multiply 10 to itself 2 times , i.e. 10 x 10 (100)

Each square has a volume of 0,1 uL = 0,0001 mL (you are calculating cell/mL). Instead of dividing by 10^-4, you multiply by 10^4 (it's the same)

We need 1000uL to have 1ml. To get this multiple 10^4 to 0.1 uL to get 1000uL. Now, to solve how many cells are in 1mL. You have to remember that solution had been diluted by a factor of 10, and also we have 10 cells per 0.1uL. if the count is 50 (avf of four) then multiply by 10 is 500 so we have 500 cells per uL
